| Objective:To investigate the correlation between the aromatic antiepileptic drug(AED)induced cutaneous adverse drug reactions(cADR) and HLA-B*1502gene in Hanpopulation of Jiangxi province.Methods:We take the case-control study,collected cases who have a cADR caused byaromatic AEDs from the neurology outpatient and inpatient of Jiangxi Children’sHospital during July2012to December2013.A total of46cases of samples werecollected,including13patients with Stevens-Johnson syndrome(SJS)(SJS group),19patients with mild skin reaction(MPE)(MPE group),and14tolerance and normalvolunteers as controls (control group).Using sequence-specific primers polymerasechain reaction (SSP-PCR) technique to detect HLA-B*1502genotype.Using Fisher’sexact test to analyze the correlation between HLA-B*1502andAED-cADRs.Results:The gene frequency of HLA-B*1502respectively are SJS group100%(13/13),MPE group10.5%(2/19), the control group7.1%(1/14). One group of data wascompared with the other groups by Fisher’s exact test. The P value both were <0.05,when the data of SJS group compared with MPE group and the control group,thissuggests that the difference was statistically significant.It is suggests that the genefrequency of HLA-B*1502of SJS group was significantly higher than that of bothMPE group and control group;However,there was no statistically significantdifference when MPE group compared with the control group(P value>0.05).Conclusion:There was a strong correlation between HLA-B*1502gene and the aromaticAEDs-induced SJS,but not MPE,in Han population of Jiangxi province. HLA-B*1502genotype testing before initiating AEDs therapy is suggested in Hanpopulation of Jiangxi province.Those with positive HLA-B*1502should avoid taking aromatic AEDs.Furthermore,physicians should also be vigilant about SCR,such asSJS,in those with negative HLA-B*1502. |
