Objective To observe the effect of traditional medicine to the expression of VEGF,and explore the intervention effect to the pathological myopia withchoroidal neovascularization (CNV) through experimental model oflasers-induced CNV in Guinea Pigs with pathologi-cal myopia inducedby form deprivation.Methods Fifteen of the1053-week old female guinea pigs were randomly selected asthe blank control group,The last guinea pigs were using facemask toinduce high myopia in4weeks, then randomly assigned to model groupand treatment group,then induced CNV by different wavelength laser.Thetreatment group gavaged with Modified Zhu Jing prescriptions while thecontrol group was gavaged with distill water.5of the blank control groupand15of the modell group and the treatment group were sacrificedimmediately on the day7,14and21,then histopathologic choroidal flatmounts, HE stain,Immunofluorescence stain, immunohistochemistry andRT-PCR in the experimental eyes and control eyes were performed.Results1.The model group and the treatment group were successfully inducedhigh myopia,and the axial length of the right eyes were significantlygreater than the left eyes. The flat mount of choroidal neovascularizationshowed that the CNV were found at the7-day,and had the biggestpermeation area at the14-day than that of the21-day in the right eyes ofthe model group. And the area of the right eyes were higher than the lefteyes.The expression of VEGF in the retinal and choroid of the modelgroup is significantly higer at the14-day than the other time detected byImmunofluorescence stain.The expression of VEGF of the treatmentgroup is less than the model group.2.The flat mount of CNV showed that the permeation area of CNV in the right eye of the treatment group were smaller than the model group and theat the14-day and21-day; the expression of the VEGF was less than themodel group.The AOD of the form deprivation eye in the treatment groupwas higher than than that in the model group except the7-day.TheRT-PCR also showed the VEGF mRNA of the treatment group was lessthan the model group.Conclusion Modified Zhu Jing prescriptions has an positive effect to the reduction ofthe VEGF expression in the choroidal neovascularization withpathological myopia and thus it can inhibit CNV.
