Objective:1、To perfect the method of digestion, cultivate and appraisal of original generation umbilical vein endothelial cells in vitro; To observe the role of the uric acid with different concentrations and last different times in the expression of vascular endothelial cells association of inflammation factors. And clear that weather the time and the concentration have any influence on the cardiovascular system.2、To visualize in a gout patient, using four different imaging techniques, the locations, sizes, compactness of tophi.Methods:1、The umbilical vein endothelial cells were separated use of0.25%trypsin and0.02%EDTA mixed digestive ferment, and cultured in the box with37℃and5%CO2in vitro Use the0.25%trypsin and0.02%EDTA mixed digest fluid to passsge. According to the unique morphological character of the human umbilical vein endothelial cells and with flow cytometric to identify the cells. Taking the vein endothelial cells in newborns umbilical, and then use the uric acid solution of different concentrations, such as0mg/dl (control group),40mg/L (group1),60mg/L (group2),80mg/L (group3),100mg/L (group4) to stimulate the venous endothelial cells for12hours,24hours,48hours and72hours. Then we use the ELISA to measure the expression of interleukin-1(IL-1), interleukin-6(IL-6), platelet activate factor (PAF) in fluid of liquid supernatant.2、The feet and knees were imaged by X-ray, MRI, DECT and PET.Results:1、①Observed in microscope, monolayer vein endothelial cells are characterized as "cobblestone", the confluent chimeric cells arranged to each other. Identified by the flow cytometric, the CD34positive rate was75.50%.②With difference concentrations in the same time:The group1has a low expression of the inflammation factors. The experimental group compared with the control group has significantly differences. P<0.05; The inflammation factor expression increased with the concentration increasing. With the same concentration in different times:In the0to12hours the expression of inflammation factor is low. The24to48hours, the expression of the inflammation factor is increased. And to the point of48hour, the expression has reached to the peak. From the48hour to the72hour, the expression of the factor has a reduce trend, but no obvious difference. It may be caused by that the test time is shorter than the time that it have the obvious difference.2、DECT uncovered deposits in unexpected locations. The combination of DECT, MRI and PET showed that tophi were much larger and more inflammatory than clinically suspected.Conclusion:1、Hyperuricemia is a risk factor to the cardiovascular disease. And the concentration of the inflammation factors relased by the vascular endothelial cell is relative to the uric acid concentrations and the action times.2、The locations and sizes of tophi in chronic gout patients can far exceed those accessible by physical examination and X-ray. These can be critical when making decisions concerning the initiation and aggressiveness of using serum uric acid-lowering therapies.
