Clinical Review Of Miao Medicine Gout And Stop Treatment Of Gout And Its Effect On HUAT And HURAT1 Gene Expression In Human Renal Tubular Epithelial Cells

Objective:1.Retrospective analysis of the therapeutic effect of Miao nationality’s herb Tongfengting treatment of damp-heat accumulation type of gout.2. To investigate the effects with different concentrations of rabbit serum containing Miao nationality’s herb Tongfengting influence on hUAT.hURAT1 mRNA expression of renal tubular epithelial cells, explore the mechanism of lowering blood uric acid,and provide a theoretical basis for Miao nationality’s herb Tongfengting treatment of gout.Methods:1.Retrospective analysis of gout patients from 2012.2 to 2014.2 in the The Second affiliated Hospital of Guiyang College who oral administration of Miao nationality’s herb Tongfengting, all syndromes type is damp-heat accumulation.A total of 85 patients was treated,in which 57 patients take the Miao nationality’s herb Tongfengting brought into the treatment group,28 patients don’t take the Miao nationality’s herb Tongfengting brought into control group, record joint symptoms,uric acid,inflammatory biomarkers and the TCM symptom score, the results are analyzed by SPSS 21.0.2.Establish renal tubular epithelial cells model, that divided into Miao nationality’s herb Tongfengting high dose group,middle dose group,low-dose group and Benzbromarone group, saline group, and intervent the cells with high,medium and low-dose rabbit serum containing,benzbromarone rabbit serum containing^ saline rabbit serum containing. The above cells were cultured in different culture medium 48 hours respectively,using real-time quantitative PCR detecting the relative expression of HK-2 cells hUAT, hURAT1 mRNA(Double standard curve method).Results:1. Retrospective study:Treatment group compared with the control group has obvious advantages(P< 0.05),and will not damage the liver and kidney function,in improving the number of patients with joint pain,tender joint count,swollen joint count,VAS pain score,reduce the TCM symptom score,ESR,CRP (P<0.05).2.High, middle and low dose groups of Miao nationality’s herb Tongfengting compared with the saline group, the hUAT expression is increased and the statistical analysis showed significantly different (P< 0.05); High,middle and low dose groups of Miao nationality’s herb Tongfengting compared with Benzbromarone group,hUAT expression increased and the statistical analysis showed significantly different (P< 0.05). High, middle and low dose, groups of Miao nationality’s herb Tongfengting compared with saline group,hURATl expression is decreased with a dose-dependen and the statistical analysis showed significantly different (P< 0.05); High,middle and low dose groups of Miao nationality’s herb Tongfengting compared with Benzbromarone group,hURATl expression levels is decreased and the statistical analysis showed significantly different (P<0.05).Conclusion:1. Miao nationality’s herb Tongfengting can improve joint symptoms、laboratory parameters and TCM symptom score for damp-heat accumulation type of gout,and will not damage the liver and kidney function.2. Miao nationality’s herb Tongfengting played a role in the regulation of HK-2 cell mRNA, upregulation of hUAT,down-regulation of hUART1,this which may be one of the mechanisms of Miao nationality’s herb Tongfengting for reducing uric acid.
