Fritillaria taipaiensis P.Y.Li (Liliaceae) is a commonly used medicinal herb in China and is recorded in the2010edition of Chinese Pharmacopoeia as the plant sources for the bulbus Fritillaria cirrhosae (Chuanbeimu). Its dried bulbs are of the functions of eliminating phlegm and relieving cough, clearing heat and moistening lung etc. Fritillaria taipaiensis is mainly directed towards symptoms like Cough with little phlegm, Expectoration with blood, cough as the reason of Yin deficiency; which is widely used as the ingredient of Chinese medicinal formulae and Chinese patent drug.This study is to establish methods to determine the ingredients in F. taipaiensis such as alkaloids and nucleotides, using the HPLC-ELSD〠HPLC-DAD from different growing periods, collecting periods and habitats. Combining with the pharmacodynamic experimental data of antitussive, expectorant and anti-inflammatory, the quality atandard of herbal F. taipaiensis is formulated. This paper can provide a sound and controlled quality standard for the construction of GAP base of F. taipaiensis.Main results of this thesis as below:1. A method that determined many ingredients (peiminine, sipeimine, peimine, peiminine) at the same time was established using HPLC-ELSD. The results showed that:content of peiminine and sipeimine was becoming less as the growing period grows. Content of peimine and peiminine was of the similar variation tendency. The comparison to the alkaloid of different processing methods(dried, well dried with mixed pulverized lime and40°C dried) showed that the content of4alkaloids in40°C dried is higher that that of the other two methods.2. The method to measure the9ingredients (nucleotide and basic group) of F. taipaiensis at the same time was established by using HPLC-DAD. The result showed that the water-soluble components of UV absorption in F. taipaiensis was ucleosides, Uracil, uridine, adenosine, adenine, inosine, thymine, cytidine, guanosine and thymidine were detected on the24collected samples. Among the detected components, the contents of uridine, guanosine and adenosine were comparatively higher.3. The fingerprint of the total alkaloid in F. taipaiensis was established using HPLC-ELSD chromatography method. The common peaks to the total alkaloid extract were confirmed on the base of its relative retention time and stability of peaks, some of which were obtained by comparing the fingerprint of chemical references. The reference peaks were achieved by using "the similarity evaluation system of TCM chromatographic fingerprint". The similarity was calculated according to these. The result showed that2samples were lower than the referencepeaks (lower than0.9), the others are higher than0.9.4. On the basis of appendix items in the Chinese Pharmacopoeia (2010)----total ash, acid insoluble ash, extract determination method, and through the survey and evaluate of the Moisture, total ash, acid insoluble ash and extracts of the F. taipaiensis among2-5years growing age and different habitats were detected. According to the study results, the suggestions were given that the moistureshould not more than17.0%, the total ash content not more than5.0%, the acid-insoluble ash not more than2.0%, extract not less than5.0%.5. The effect of cough caused by ammonia was studied by using the powder and alcohol extract of F. cirrhosa, hydrobromide right Suofen as positive control, and powder and alcohol extract of F. taipaiensis as test samples. The Tracheal phenol red excretion of mice was also studied by using the powder and alcohol extract of F. cirrhosa,. and guaiacol glyceryl as positive control, nd powder and alcohol extract of F. taipaiensis as test samples. The resulted that powder, alcohol extract of F. taipaiensis was of the similar effects as these of bulbus F. cirrhosae, which has significant antitussive, expectorant and anti-inflammatory effect.6. The quality standard of F. taipaiensis was drafted according to its difference of component contents and fingerprint from different growing periods and habitats, and the experimental data of powder and alcohol extract. |
