Background and ObjectiveCervical cancer is one of the main causes of the death of women’s cancer and a public health problem in all over the world.The Morbidity and mortality increased gradually in recent years.The data of recently show that, cervical cancer is closely related to human papillomavirus (HPV).However,the virus can be removed in most people of HPV infection in12-30months.So, it is supposed that,cervical cancer besides HPV infection is also related to other factors, especially the organism immune state. There are many types of immunocytes in human body,like macrophages,NK cells and B lymphocytes.In normal condition, they interact to each other to maintain the symmetry of immunity system. When the number and function of these immunocytes change, it may lead to the development cervical cancer.Formerly, the research of macrophages,NK cells and the B lymphocytes mainly concentrated in peripheral blood in cervical cancer patient’s.There were rare reports about immunocytes in cervical cancer tissue and also rare reports about the relationship between the immunocytes and HPV. In this study, we quantified the number of macrophages,NK cells and the B lymphocytes in cervical cancer by immunohistochemical staining, and analyzed the relationship between these immunocytes and HPV infection and the role of these immunocytes in development of cervical cancer.Materials and Methods1. We collected146cases of paraffin-embedded tissues of cervical lesions that included20cases of cervicitis,9cases of low grade CIN (CIN1),34cases of high grade CIN (included16cases of CIN2and18cases of CIN3),59cases of cervical squamous cell carcinoma (SCC)and24cases of cervical adenocarcinoma(ADC).2. HPV genotyping in all cases was detected by HPV DNA-chips technology.3.Immunohistochemistry was used to detect macrophage,NK cells and the B lymphocytes.Results1.The number of macrophages was7.17±3.03in cervicitis group,8.87±0.86in low grade CIN,8.50±0.40in high grade CIN,9.70±2.86in SCC and12.46±8.47in ADC. The number of macrophages in SCC and ADC group was significantly higher than cervicitis,low grad CIN and high grade CIN (P<0.05)2.The number of NK cells was26.34±15.19in cervicitis,21.00±2.02in low grade CIN,14.11±0.95in high grade CIN,13.05±7.03in SCC and12.40±6.79in ADC. The number of NK cells in high grade CIN, SCC and ADC was significantly lower than cervicitis and low grade CIN (P<0.05).3. The number of B lymphocytes was32.34±22.57in cervicitis,29.72±3.36in low grade CIN,27.88±5.79in high grade CIN,71.38±37.33in SCC and52.58±30.61in ADC. The number of B lymphocytes in cervicitis,low grade CIN and high grade CINwas significantly lower than SCC and ADC (P<0.05).4. In146cases,there were116cases of HPV-positive and,.The HPV infection rate leaded up to79.45%.Among HPV positive116cases,54cases were High-risk HPV16positive,19cases were HPV18positive,21cases were infected by the following subtypes:HPV11,31,33,35,42,45,51,52,56,58,59,66); and16cases were infected by the mixed infection (included HPV HPV6.16;16.18;11.16;16.31;16.33;16.52;16.59;16.58;16.68;18.16;31.58;58.6;52.33;16.33.59)5. The relationship between macrophages,NK cells,B lymphocytes of each group and HPV infection. In cervicitis group,the number of B lymphocytes was38.89±4.92in HPV-negative cases,which were higher than12.68±0.07in HPV-positive cases(P<0.05).In SCC group,the number of B lymphocytes was46.50±2.90in HPV-negative cases,which were lower than72.26±4.99in HPV-positive cases(P<0.05).In the other groups,there is no significant difference between each other in HPV-negative cases and HPV-positive cases(P>0.05).6. The relationship between all cases and HPV infection.In HPV-negative cases, the number of NK cell was18.56±1.93,which were higher than14.71±0.83in HPV-positive cases(P<0.05).In the other groups,there is no difference between each other in HPV-negative cases and HPV-positive cases(P>0.05).Conclusions1.Compared to cervicitis and CIN, the number of macrophages in cervical cancer tissue were increased.2. NK cells in cervical cancer have a lower number than in cervicitis, From cervicitis, CIN to cervical cancer,the number of NK cells reduce gradually.3. In cervical, B lymphocytes have a higher number than in cervicitis,and CIN.But the effect of B lymphocytes is uncertain.4.HPV infection changed number of B lymphocytes in cervicitis and SCC,and had small impact on macrophages and NK cells.HPV infection also made a little change of these three types of immunocytes in low grade CIN,high grade CIN and ADC.But in all HPV infection cases,the number of NK cells changed,it reflect that HPV infection to restraint of nonspecific immunity,and promote the process of disease.
