| Saussurea involucrata (Kar.et Kir.) belongs to the genus Saussurea (Composite), habitually used by Uygur ethnic group. It has the property and flavor with muggy, mild bitter and warm, effects in bushenhuoxue and humoral regulation, strengthening the bones and muscles, trophic nerve, used in the therapy of malvarmumo, arthritis deformans and gynecological disease. Due to its special medical values, unrestrained exploitation causes the decrease of resources and eventually is catalogued as the endangered space by the nation. As the relative preparations from various sources launched in the market, the Chinese Pharmacopeia (2010 ed) defines methods using rutin and chlorogenic acid as standards to identify and analysis its quality, but this prone to induce the addition of standards artificially. We intent to establish the analysis system of fingerpints based on biological and chemical studies of Saussurea involucrata (Kar.et Kir.)The 65%EtOH extract of S. involucrata were extracted with EtOAc and BuOH and the isolation and purification of compounds from these two parts are carried out.13 compounds were identified by various spectrum methods and it reviewed that 3 of them are flavones,7 of them are guaiane sesquiterpenes which was characterized by 11βH-11,13-Dihydrodehydroco-stuslactone 8α-O-β-D-glucopyranoside (10).HPLC and TLC methods were investigated on the fingerpintss of Saussurea involucrata. The HPLC fingerpintss of Saussurea involucrata was established by EtOAc extract. The chromatographic conditions were determined by mobile phase:water (A)-acetonitrile (B), using a gradient program of 8%B-100%B (0-75min) and by detection wavelength at 200 nm.10 peaks were designated and 7 of them were identified as:costic acid (1); 8a-Hydroxy,11βH-11,13-dihydrodehydrocostuslactone (3); 3α,8a-dihydroxy,11βH-11,13-di hydrodehydrocostuslactone (6); 11β,13-dihydrodeacylcynaropicrin (7); 11βH-11,13-dihydro dehydrocostuslactone 8α-O-β-D-glucopyranoside (10); 11β,13-dihydrodesacylcynaropicrin 8-β-D-glucoside (12); rutin (13). Medicinal plants extract from four habitants, along with semi-product and preparations (oral liquid from three lots) were tested based on the fingerpints. The result showed the difference between habitants, and the similarity of medical plants extract and semi-product, it also discovered that the main peak which existed in two of oral liquid didn’t show in other samples, and it remains to be tested in further study. TLC fingerpintss of Saussurea involucrata showed better in 365 nm wavelength than in visible light. The result showed the similar difference between habitants with that in HPLC method. The way of difference between oral liquid and medicinal plants extract were similar with their HPLC behavior, but the oral liquid from three lots existed not many differences between them which showed differently by their HPLC method.The hydrophobic constituents in Saussurea involucrata from 4 habitats were extracted with chloroform and 56 chemical components was identified by GC-MS method. The hydrophobic constituents from 4 different habitats were calculated with relative content. They were mainly classified to unsaturated aliphatics, saturated aliphatics, sesquiterpenoids, steroids and triterpenoids, consisting the most abundant constituents including n-hexadecanoic acid; 6-(1-hydroxymethylvinyl)-4,8a-dimethyl-3,5,6,7,8,8a-hexahydro-1H-na phthalen-2-one; 4,7,10,13,16,19-docosahexaenoic acid, methyl ester (all-Z)-; azuleno[4,5-b] furan-2,9-dione,9a-[(acetyloxy)methyl]decahydro-6-methyl-3methylene-,[3aS-(3α,6β,6aa,9aβ,9bα)]-; 9,12-octadecadienoic acid (Z,Z)-; 9,12,15-octadecadienoic acid (Z,Z,Z)-; nonacosane; hentriacontane;β-sitosterol and a-amyrin, some individual components vary significantly among hydrophobic constituents from different habitats.4,7,10,13,16,19-docosahexaenoic acid, methylester (all-Z)-; azuleno[4,5-b]furan-2,9-dione,9a-[(acetyloxy)methyl]decahydro-6-methyl-3 methylene-,[3aS-(3aα,6β,6aa,9aβ,9bα)]-; 9,12-octadecadienoic acid (Z,Z)-and 9,12,15-octadecadienoic acid (Z,Z,Z)-were first discovered in hydrophobic constituents of Saussurea involucrata.In the anti-bacteria activity of the hydrophobic constituents from four habitants,96-well plate were used to test its minimum inhibitory concentration (MIC) on S. aureus and its 5 multidrug resistant strains for evaluating its anti-bacteria activity. The result turned out to be no obvious activity among four habitants. The hydrophobic constituents from four habitants and other different polarity sections from extact of Saussurea involucrata were examined for antioxidant activity. The result showed minor difference on the clearance of DPPH between four habitants. Compared with Vitamin C and Vitamin E, the hydrophobic constituents didn’t show significant antioxidant activity, but the clearance of DPPH of hydrophobic constituents increases as the concentration goes. At the concentration of 2 mg/mL, the clearance of DPPH is 25%-45%. This is the first time to test the antioxidant ability of different polarity section from extact of Saussurea involucrata and the results reviewed that the antioxidant ability of extract from EtOAc and BuOH are higher than the total extract and the remaining section, and the EtOAc extract showed the highest:at the concentration of 0.2 mg/mL, the clearance of DPPH reached equilibrium (93%). |
