Saussurea involucrata is one of the most famous and important medicinal herbs in China.In this study, various chromatographic techniques were employed to isolate and analysis chemical constituents in the 95% ethanol extraction of the cultures of S. involucrata. Fifteen compounds including one new compound (8) and fourteen known compounds were characterized by the analysis of spectroscopic data. They are palmitic acid(1),β-sitosterol (2), (2S,3S,4R)-2-tetracosanoylamino-1,3,4-octadecanetriol (3), hexacosanoic acid (4), linoleic acid (5),3-O-(6’-O-linoleoyl-β-D-glucosyl)-β-sitosterols (6), linoleic acid monoacylglycerol (7), 11βH-2α-hydroxy-eudesman-4(15)-en-12,8β-olide (8),3-O-(6’-O-Palmitoyl-β-D-glucosyl)-β-sitosterols (9), (2S,3S,4R,11E)-1,3,4-trihydroxy-2-[(2’R)-2’-hydroxytetracosanoylamino]-11-octadecene (10),β-daucosterols (11), syringin (12), benzyl alcohol glycoside (13), tangshenosideⅢ(14) and 1,5-di-O-caffeoylquinic acid (15). The yield of syringin is about 0.3% of the dry weight of cell cultures. The pharmacological activities of syringin were evaluated.A quantitative analytical method for the content of syringin in cultures of S. involucrata was established by RP-HPLC. The method is convenient, accurate and reliable. All this results provided a basis for further studies on application of cell culture of S. involucrata for large-scale production of bioactive compound syringin. The contents of syringin in wild S. involucrata medicinal materials and in different optimized cell culture conditions were determined. The contents of syringin in wild plant, callus, cultures from cell suspension culture and bioreactor culture are 0.027%、0.317%、3.289% and 1.086% respectively. Besides, under the best conditions, the content of syringin in cell suspension culture is about 120 times of the content in wild S. involucrata medicinal material, and the yield of syringin can reach 740 mg per liter.
