Research On The Influence Of Fusarium Toxin ZEA And DON On The Function Of Mouse’s Humoral Immunity And Antioxidant Function

180 female healthy Kunming mice were chosen as samples of our experiment in accordance with the rules of 2 elements and 3 standards. The respective consistency of ZEA is 0 mg/kg.BW、20 mg/kg.BW、30 mg/kg.BW,while the consistency of DON is 0 mg/kgB.W、1.5 mg/kgB.W、2.5 mg/kgB.W. These mice were divided into 9 groups in random including 8 for experiment and 1 for comparison. All the mice were continuously injected for 4 times in abdominal cavity every 24 hours with different consistency of DON and ZEA. After the first injections, We got 4 samples respectively from all goups in the 3d(before the third injection)、the 5d、the 8d as well as the 12d. We collected blood from decapitation and made it into serum. According to the data of immune globulin (IgA, IgQ IgM)、 levels of interleukin(IL-1, IL-4)、 level of complement 3、activity of superoxide dismutase (SOD)、malonaldehyde (MDA) and hydroxy free radical (OH-), which were got from the serum. From those data,we wanted to evaluate the mixture and cumulative immunotoxicity and the influence of antioxidant function of ZEA and/or DON.The conclusions were:(1) During exposure process, the in vivo effects of the treatment showed that serum IgA levels of group D2 (DON 2.5mg/kg) were significantly lower (P< 0.01) than the control group and before exposure, while other groups were significantly higher (P< 0.05 or 0.01) than the control group; The serum IgG levels of all groups were significantly higher (P< 0.05 or 0.01) than the control group and before exposure, except that the group D2 was significantly lower (P< 0.05 or 0.01) than the control group and before exposure on the fifth day; The serum IgM levels of all groups were siginificantly lower (P< 0.05 or 0.01) than the control group and before exposure on the fifth day, except group D1; ZEA has no dose-effect influence on IgM, while DON and ZEA have obvious dose-effect influence on immune globulin; Mixture exposure with high concentrations of ZEA and DON show obvious additive effect, especially the serum immune globulin levels of group D2Z2 (DON2.5mg/kg+ZEA30mg/kg) were significant different (P< 0.01 or 0.05) at each time point with its single exposure group. (2) DON and ZEA could make IL-1 and IL-4 dysregulation, the exposure groups significantly improved the serum IL-1 and IL-4 of mice, except that group D2 declined the serum IL-1 and IL-4 obviously and group Z1 had no significant impact at the third day; ZEA and DON showed obvious dose-effect influence on serum IL-1 and IL-4 of mice; Combined exposure with ZEA and DON showed obvious additive effect, the mixture groups were significant different (P< 0.01 or 0.05) at each time point with its single exposure group, except that the IL-1 levels of samples took at the eighth day of group D1Z2, the IL-4 levels of samples took at the fifth, eighth day of group D1Z1 and the IL-4 levels of samples took at the 12th day of group D1Z2; (3) During exposure process (The third and fifth day), the exposure groups significantly improved (P<0.01) the serum C3 levels, except that the group D2 significantly declined the C3 levels; Our experiments showed that only DON showed obvious dose-effect influence on serum C3 levels; Mixture exposure with ZEA and DON showed obvious additive effect, the combined groups were significant different (P<0.01 or 0.05) at each time point with its single exposure group, except that the samples took at the third day of group D1Z1, the samples took at the eighth,12th day of group D1Z2 and group D2Z1. (4) DON and ZEA significantly improved (P<0.01)of the serum MDA levels and reduce the serum inhibiting capacity to OH- activity of mice, that was observed at all the exposure groups; The single exposure of DON and ZEA could improve the activity of the SOD, except that the group D2 significantly declined (P<0.01) the activity of the SOD; Mixture exposure with ZEA and DON showed obvious additive effect on peroxidation products, especially the MDA and OH- levels of group D2Z1 and group D2Z2 were significant different from its single exposure groups (P<0.01 or P<0.05) at each time point; All the activity of the SOD of mixture exposure groups were significant different (P< 0.01 or P<0.05) at each time point with its single exposure group, excpet the group D1Z2, the results showed that mixture exposure has obvious additive effect on the activity of the SOD. (5) The immune function and antioxidant function of group D2、group D2Z1 and group D2Z2 had been in an abnormal state even after exposure, while the indicators of other groups had the trend to approach to the control group and before exposure.The results showed that:ZEA and DON has negative impact on humoral immune through causing immunoglobulins, interleukins and complement dysregulation, at the same time they have bad influence on the body antioxidant function, that’s also will have indirectly affect on the humoral immune. The mixture exposure will exacerbate the toxicity, especially at the high concentration of mixture exposure, the additive effect would be more obvious.
