| Indigofera Kirilowii Maxim. ex Palibin is a native tree species with important medicinal and landscaping value in the north. Now studies is only limited in the field of introduction and medicine. There is no reports about the genetic diversity and the core collection in I. Kirilowii Maxim. ex Palibin. In this study, genetic diversity, genetic structure and core collection was analysed by AFLP markers in order to valuation, protection, development and utilization of I. Kirilowii Maxim. ex Palibin. The result as follow:1. Genetic diversity of I. Kirilowii Maxim. ex Palibin: 8 primer combinations selected from 64 primer combinations produced a total of 1 589 bands, 1587 of which were polymorphic with a polymorphism percentage of 99.87 %. The value of Na, Ne, H, I were 1.9988, 1.2157, 0.1401 and 0.2346, which indicated that I. Kirilowii Maxim. ex Palibin had a high genetic diversity. 139 specific bands including 1 absent band were amplified from 220 single plants, which account for 8.75 % of total polymorphic bands. Genetic similarity coefficient of 220 single plants changed from 0.1884 to 0.8328 with the average was 0.5528. In the clustering results, the germplasm from Jinan, Qingdao, Weihai, Yantai and Zibo were closed, and which from Linyi, Rizhao and Taian dispersed. Ploymorphism percentage of 8 populations changed from 43.05 % to 75.05 % with the average was 58.25 %. The order of polymorphism percentage from big to small was: Taian>Yantai> Linyi> Weihai> Jinan> Zibo> Rizhao> Qingdao.2. Genetic structure of I. Kirilowii Maxim. ex Palibin: Gst was 0.1058, it suggested genetic variation was mainly within the populations, Nm was 4.2904. Genetic identity ranged from 0.9750 to 0.9932 with the average was 0.9844. Genetic distance changed from 0.0068 to 0.0254 with the average was 0.0158. All the populations were clustered into 4 groups at 0.014, and that there was no strict relevance between the genetic and geographic origin of the I. Kirilowii Maxim. ex Palibin.3. Core collection of I. Kirilowii Maxim. ex Palibin: The core collection was built as proportion sample of 30 %, which including 66 germplasm. The reserved rate of number of polymorphic loci and percentage of polymorphic loci were 93.43 % and 99.87 %, and the reserved rate of observed number of Na, Ne, H, I were 99.93 %, 101.23 %, 107.99 % and 108.70 % respectively. Genetic similarity coefficient of core collection plants changed from 0.1879 to 0.6427 with the average was 0.5356, and all the populations were clustered into 9 groups at 0.53. The finger-print of I. Kirilowii Maxim. ex Palibin was built by AFLP result. |
