| Chinese yellow cattles have some defects such as small physique, Slow growth,hindquarters underdeveloped (low meat yield).However, meat production and meat qualitytraits is a typical minor quantitative trait controlled by multiple genes,Therefore, theheritability relatively low,If using traditional phenotype breeding methods will encounter lowselection efficiency, relatively poor accuracy, long generation interval, genetic progress moreslowly and other issues. In this study,use silence adjustment factor Sirt3gene and Sirt5genewhich closely related of energy and lipid metabolism in Qinchuan cattle for the object, theaim of the study was to research polymorphism of Sirt3gene and Sirt5gene and it associationwith Body Measurement Traits and Meat Traits in Qinchuan Cattle, then finding molecularbreeding methods of improving Body Measurement Traits and meat traits in Qinchuancattle.In this study, Qinchuan heifers from18-24month old under similar feeding conditionswere randomly selected. Direct DNA sequencing methods were used to genotype thepolymorphisms in Sirt3gene(468) and Sirt5gene(572). Least squares fitting linear modle ofSPSS16.0program were used to analyze the relationship between the different genotypes andBody Measurement Traits and Meat Traits. The aim was to choose perfect markers which canprovide theoretical base for breeding in Chinese cattle. The results were as follows:Two sites existed polymorphism in Sirt3gene and four sites in Sirt5gene,which wereexon5and intron5in Sirt3gene and3’UTR in Sirt5gene. The analysis of associationbetween different genotypes and body measurement traits and meat traits were showed asfollows:1The polymorphism analysis of the exon5and intron5of Sirt3geneTwo SNPs had been detected in Sirt3gene including C22522T in exon5, C22680G inintron5.The result of direct DNA sequencing methods showed that C22522T and C22680Gshowed three genotypes. Test showed C22522T and C22680G were highly not inHardy-Weinberg equilibrium(P<0.01). Association of Sirt3Gene Polymorphism with BodyMeasurement Traits and Meat Traits showed that the genotypes of different mutation in Sirt3 gene were significantly related to body length, withers height, hip height, rump length, pinbone width and back-fat thickness, lion muscle area, intramuscular fatty content (P<0.05orP<0.01). It indicated that SNPs of the Sirt3gene may be closely associated with some BodyMeasurement Traits and Meat Traits, and could be a major candidate gene for new strainbreeding of Qinchuan cattle.2The polymorphism analysis of the3’UTR of Sirt5geneFour SNPs had been detected in Sirt5gene including G22010A, G22052A, G22119T andG22245C in3’UTR, The result of direct DNA sequencing methods showed that G22010A andG22052A showed three genotypes, G22119T and G22245C showed two genotypes. Testshowed G22010A,G22245C,G22119T were not in Hardy-Weinberg equilibrium(P<0.05),G22052A is highly not in Hardy-Weinberg equilibrium(P<0.01). Association of Sirt5GenePolymorphism with Body Measurement Traits and Meat Traits showed that the genotypes ofdifferent mutation in Sirt5gene were significantly related to body length, withers height, hipheight, rump length, pin bone width and back-fat thickness, lion muscle area, intramuscularfatty content (P<0.05or P<0.01). It indicated that SNPs of the Sirt5gene may be closelyassociated with some Body Measurement Traits and Meat Traits, and could be a majorcandidate gene for new strain breeding of Qinchuan cattle. |
