Polymorphisms in GH-P1 locus and their association with partial body measurement traits in Qinchuan cattleThe research was conducted to detect SNPs in GH gene P1 locus of Qinchuan cattle. Results showed that a C→G mutation was founded in the 4th base in exon 5 of GH gene ,which led the amino acid changing from Leu to Val. There were 3 genotypes named AA,AB and BB respectively after digestion by Aluâ… .Allele frequency of A and B were 0.825 7 and 0.174 3 respectively.Χ2 test showed that the locus was not in Hardy -Weinberg equlibrium in the Qinchuan cattle population detected. Correlation analysis showed: individuals with different genotypes had significant differences in wither height, heart girth, body length and rump length.2. Polymorphisms in GH-P2 locus and their association with partial body measurement traits in Qinchuan cattle. The study was to detect SNPs in GH gene P2 locus of Qinchuan cattle. The result showed that a C→T mutation was founded in the 93th base in exon 5 of GH gene, which was synonymous mutation. There were 2 genotypes named CC and CD respectively, after digestion by Aflâ…¡.Allele frequency of C and D were 0.902 4 and 0.097 6 respectively.Χ2 test showed that the locus wasn't in Hardy-Weinberg equlibrium in the Qinchuan cattle population detected.Correlation analysis showed: individuals with different genotypes had significant differences in hip width, ischium width and rump length. 3. Combined genotypes in P1 and P2 locus in GH exon 5 and their relationship with partial body measurement traits in Qinchuan cattleThe result of analysis of two loci combined genotypes in exon 5 showed that body measurement traits of combined genotype ABCD in exon 5 were higher than AB and CD genotype and other kinds of combinations genotype. The effects of multi-locus combinations of marker were higher than those of single-marker locus.Combined genotype ABCD may be the best genotype combination in affecting body measurement traits of Qinchuan cattle.4. Polymorphisms in GHR-8 locus of GHR gene and their association with partial body measurement traits in Qinchuan cattleThe study was to detect SNPs in GHR-8 locus of Qinchuan cattle. The result showed that a T→A mutation was founded in the 4962th base in GHR gene ,which led the amino acid changing from Phe to Tyr. There were 3 genotypes named EE,EF and FF respectively after PCR-SSCP.Allele frequency of E and F were 0.610 6 and 0.389 4 respectively.Χ2 test showed the locus was not in Hardy -Weinberg equlibrium in the Qinchuan cattle population detected. Correlation analysis showed: individuals with different genotypes had significant differences in heart girth. In the other body measurement traits , individuals with EF and FF genotypes were higher than the EE genotype ,but no significant difference. | | Keywords/Search Tags: | Qinchuan cattle, Growth Hormone gene, GHR gene, PCR-RFLP, PCR-SSCP, Combined genotype, Body measurement traits | PDF Full Text Request | Related items |
