Dairy cow Mastitis has been one of the most serious and unpreventable disease in dairy production. The wide application of western medicine contributes to side effects such as Antibiotic residues and antimicrobial resistance, thus the increasing proportion of relapse is yet to be alleviated.Compound medicine equipped with the advantages of lower level of poisonous effects, chemical residues and antimicrobial resistance, Mongolian veterinary compound stands out as a potential solution. This research aim to screen anti-inflammatory active composition for "special Moisten Dole" by Orthogonal method, and then test its effect in dairy cows Mastitis.The main results of this research are presented as followed:1. The optimal combination turned out to be A2B2C3D1F1G3, wich means moderate dose of A B E, high dose of C and no DFG, judging by the assessed antibacterial effects.2. Maximum tolerated dose of this compound is5g/kg·b·w. According to the provision of "Veterinary test specification compilation-new veterinary-General technical requirements for toxicity testing", the compound of A2B2C3D1E2F1G3is qualified as toxin free.3. Applied the compound to mice of Mastitis, which was exclusively infected by a mix of3pathological bacterial strains. The biopsy showed that this compound work in resisting infection, alleviating congestion and swelling in its mammary tissue, and facilitating its restoration.4. We extracted the active ingredients in geniposide, curcumin, rhein and toosendanin successfully. Their respective extracts’s proportion was73%,73%,79%,63%.5. The compound was proved to significantly promote immunity on mice:spleen index was significantly improved (P<0.05); phagocytic index K and phagocytic coefficient A were significantly improved (P<0.01);activity of NK cell was significantly improved (P<0.05); no significant effect in peripheral CD3+T cell proportion, CD4+T cell proportion, CD8+T cells proportion and CD19+B cells proportion (P>0.05); CD4+T/CD8+T ratio and CD14+cells proportion were significantly improved (P<0.05). The level of IL-2in breast tissue model mice was significantly increased (P<0.01). IFN-γ level in breast tissue model mice was significantly increased (P<0.05). IL-6, IL-8, TNF-α and NAGase elevating were significantly inhibited (P<0.05). No significant effect in IgG, IgA levels (P>0.05).It is suggested that the compound can boost non-specific immunity in mice by enhancing Mononuclear macrophages phagocytosis and NK cell activity, and specific immunity by increasing CD4+T/CD8+T ratio and CD14+cells proportion. By adjusting the amount of the secretion of cytokines such as IL-2and enzymes to improve immune and reduce inflammatory injury.6. The anti-inflammatory test on mice showed:The increasing capillary permeability induced by acetic acid was significantly inhibited (P<0.05), the swelling caused by inflammation sites was significantly inhibited (P<0.05). The elevating PGE2level from formaldehyde-induced swelling was significantly inhibited(P<0.05). The granuloma caused by cotton was significantly inhibited (P<0.01). The contents of E-selectin and ICAM-1in mouse serum were significantly reduced (P<0.01).As it indicated, the compound can regulate the inflammatory response effective, which was controlled within a proper range.
