Siraitia grosvenorri is a traditional and medical herbs of China. This paper used fresh Siraitia grosvenorri from Guangxi province as material to perform research in the follow three aspects.(1) In order to investigate effect of different drying methods on the fresh Siraitia grosvenori, blast drying, far infrared drying, microwave drying and vacuum drying were compared accorrding to the variation of moisture content and fruit quality during the drying process. Afer process optimization, microwave combined with vacuum drying performed well. By measuring the content of total mogrosides and flavonoids, the retention of active ingredients in dried fruit was studied, and the sensory evaluation criteria system were established. After microwave combined with vacuum drying, the content of active ingredients maintained well, that is, mogrosides and flavonoids content reached 1.63%, 1.33% respectively, and the dry fruit also get the highest score in sensory evaluation consist of appearance, sensory and flesh structure.(2) Effect of ethanol concentration, ratio of solvent to material, and times of extraction and ultrasonic extraction time were determined on the basis of extraction yield of total mogrosides. After consideration of practical experence and cost for control, yeild of total mogrosides reach to 3% following the extraction condition: 30% concentration ethanol aqueous, material-to-liquid ratio of 1:15(w:v), extraction 1h each time for total 3 times.(3) After extraction from dry fruit and purification by chromatography column of silica gel, LH-20, MCI, D101 resin, ODS and semi preparative HPLC, 7 mogroside compounds were isolated and 6 of which were identified. They are Mogroside IV, Mogroside IIIE, siamenoside I, Mogroside IVa, Mogroside V and Iso mogroside IV.
