Objective:To investigate the intervention of Yiqiqingre cream and Dioscin (The important active ingredient of traditional Chinese medicine Chuanshan Long) on rat mesangial cells(MC) proliferation and to explore the underlying mechanism.Methods:1. The first experiment:Research by serum pharmacology method. Wistar rats were fed with Yiqiqingre cream in order to make drug-containing serum, and drug-containing serum was added into the vitro experiments. MC were divided into control group, lipopolysaccharide(LPS) stimulated group, Yiqiqingre cream-contained serum group(YQ-S group) and Fosinopril-contained serum group(For-S group), and cultured48h. MTT assay was used to detect the proliferation of MC. RT-PCR and Western blot methods were applied to detect Wnt4, β-catenin and TGF-β1expression of the MC.2. The second experiment:Dioscin-the active ingredient of Dioscorea, directly was added into the vitro experimental. MC were divided into control group, lipopolysaccharide(LPS) stimulated group, LPS+Dioscin(0.1μg/ml,0.5μg/mk、1μg/ml) group, and cultured48h. MTT assay was used to detect the proliferation of MC. RT-PCR and Western blot methods were applied to detect Wnt4, β-catenin and TGF-β1expression of the MC.Results:1. The first experiment results showed Yiqiqingre cream-contained serum can inhibit LPS-induced MC proliferation. Wnt4and TGF-β1mRNA expression levels were reduced in YQ-S group(P<0.01or P<0.05). Wnt4, β-catenin and TGF-β1protein expression were suppressed in YQ-S group(P<0.01or P<0.05), Its role was similar to and the effect of the positive control drug fosinopril in the experiment.2. The second experiment results showed Dioscin can inhibit LPS-induced MC proliferation and the expression levels of Wnt4、β-catinin and TGF-β1(P<0.01or P<0.05) in a dose dependent manner.Conclusion:The two experiments showed Yiqiqingre cream and Dioscin can inhibit rat mesangial cell proliferation, its mechanism may be related to inhibition of the Wnt signaling pathway, and reducetion of TGF-β1expression, considering positive correlation between the expression of Wnt signaling pathway component and TGF-β1, We can speculate that there is some connection between the Wnt signaling pathway and TGF-β1-mediated signaling pathway. Dioscin is the active ingredient of Chuanshan Long, its role in the experiment was similar to Yiqiqingre cream, We can infer Yiqiqingre cream might play the inhibition effect of rat mesangial cell proliferation through Dioscin.
