Objective: (1)To investigate the clinical effect on reflux esophagitis of invigorating thespleen,dispersing the stagnated liver-qi, activating qi flow and calming the adverse rising qidecoction, and provide new clinical route for Chinese Medicine treatment of RE. (2) inorder to find the best way to make animal modle for RE, we contrast the different operationcharacteristic and study the mechanism of the treatment rules of "invigorating the spleen,dispersing the stagnated liver-qi, activating qi flow and calming the adverse rising qi" onRE in the respect of accommodating gastroesophageal motivation and protecting mucousmembrane, we prove active effect is lowering RE’s canceration risk by observation of theexpression of COX-2 and p53. We form a base for mechanic research and seekingfunctionary target of traditional Chinese drug on RE.Method: (1)Clinic research: 45 reflux esophagitis patients whose TCM syndrome are spleendeficiency and liver stagnation were divided into two groups randomly: treatment group are cured byChinese Medicine and positive group by western medicine. The two groups were respectively giveninvigorating the spleen,dispersing the stagnated liver-qi, activating qi flow and cahning the adverserising qi decoction and Mosapride Citrate Tablets for 4 weeks. We study the change of major andsecondary symptoms before and after the treatment. (2)Experiment study:①We establish three kindsof animal model by different operations on healthy SD rats, then we observe the every group rats’ deathrat, weight, esophageal mucous membrane.②The rat models of reflux esophagitis were established by"partly pyloric ligation+cardiomyotomy". We randomly divided them into seven groups: blankcontrolled group, model group, placebo group, western medical controlled group, and high、middle andlow dosage of Chinese medicine groups. then we study the effect of the treatment by observing thechange of Cox2、p53、GAS,etc.Result: (1)The two clinic groups all had no significant difference between major andsecondary symptoms(P>0.05). Treatment group being cured by Chinese Medicine hadsignificant change of some symptoms, such as degree of heartburn, frequency of chest pain,belch, chest pain, irritable compared with positive group. (2)The method of "partlypyloric ligation+cardiomyotomy" have the peculiarity of lower death rat (27.6%) andhigher success (77.8%). Animal research show that invigorating the spleen, dispersing the stagnated liver-qi, activating qi flow and calming the adverse rising qi decoction couldimprove pH of lower esophagus, alleviate pathology injure, strengthen the esophagusamplitude, reduce the expression of COX-2 and p53. The effect between the two groupshave no significant difference (P>0.05).Conclusion: invigorating the spleen, dispersing the stagnated liver-qi, activating qi flowand calming the adverse rising qi decoction has effect on RE. We infer the mechanism isimprove motivation, regulate inflammatory factors of mucous membrane of esophagus,restrain cancer gene, etc.
