Objective:To test the expression of EGFRvⅢ and PDGF-D in the colon carcinoma of different differentiation degree and clinical stage and normal control group. And To investigate its effections in the increment, invasion and metastasis. Meanwhile to analyse the correlation between them and to study whether they can be the consensus index of colon carcinoma prognosis.Methods:60tissue samples from patients which with radical operation for carcinoma of colon were entrolled and20normal colon tissues were chosen as control.Using immunohistochemistry method to test the expressions of EGFRvⅢ and PDGF-D and analyse the expression degree in the two groups,and to analyse the correlation of EGFRvⅢ and PDGF-D and the relationship between them and colon cancer’s differentiation degree and clinical stages.Results:1. The result of immunohistochemistry showed:EGFRv Ⅲ mainly was expressed in the cell membrane, and some cytoplasm were coloured; PDGF-D was mainly expressed in the cell membrane and cytoplasm.2.60cases of colon cancer tissues, the positive expression rate of EGFRvⅢ and PDGF-D was respectively66.67%and63.33%, which was significantly higher than that of the normal colonic mucosa tissue group with0.00%and25.00%(p< 0.01).3.The express of EGFRv Ⅲ and PDGF-D in colon cancer tissue were related with tumor differentiation. The positive expression rate of EGFRvⅢ in the high, medium and low differentiated carcinoma were respectively40%,70%and90%;the positive expression rates of PDGF-D were respectively45%,60%and85%, thus there were significant statistical differences of these groups (p<0.05).4. The positive expression of EGFRvⅢ in the stage Ⅲ and IV was75.56%, which was higher than that in the stage Ⅰ and Ⅱ with40%(p<0.05); the positive expression rate of PDGF-D in stage Ⅲ and IV was71.10%, which was higher than that in the stage Ⅰ and Ⅱ wit 40%(p<0<0.05).5.There was a close correlation between EGFRvⅢ and PDGF-D expression in colon carcinoma (r=0.636, p<0.01)ConclusionEGFRvⅢ and PDGF-D specificly expresse in colon cancer and have close relationship with pathologic differentiation degree in colon cancer.They have positive correlation, and can be the consensus index of grade malignancy assessment,progress and prognosis assessment of colon cancer.
