Objective: Intrahepatic cholangiocarcinoma (ICC), a kind of commonmalignant tumor, which is difficult in early diagnosis, low cure rate and poorprognosis, etc. Rat model was conduct in this experiment, to study theexpression of matrix metalloproteinase2,(MMP-2) and Interleukins6(IL-6).Method:60Sprague Dawley (SD) rats with half males and half femalesrange from8to12week and180to220g were used in this experiment. Theywere dividing into2groups the experimental group (40rats) and the controlgroup (20rats). The diets used in experimental group contain3`-Me-DBA,while the control group are common.9rats in the experimental group and4rats in the control group (2rats at18week) were killed at12week,14week,16week and18week, respectively. To collect the bile,liver, and observe theorgans. The liver were collect to make paraffin. Elisa was used to measure theconcentrations of MMP-2and IL-6, and HE was use to dye paraffin forobservation and analysis the correction between the two groups.Result:1.The result showed that there are significant differencesbetween the two groups. This observation prove that correspond with theprocess:Hepatitis to Liver fibrosis to Atypical hyperplasia to ICC. In thisexperiment we induced12case of cancer that contain7case of highdifferentiation,3case of middle differentiation and2case of lowdifferentiation. The rate of cancer increased greatly after16weeks.2.MMP-2,IL-6in rat bile level of Experimental group was significantly higher than the control group (P<0.05); In different stages of inducing cancer, the differenceof MMP-2, IL-6expression in bile had significant statisticalsignificance(P<0.05), expression of MMP-2increased with increasing ofinduced cancer weeks; expression of IL-6had no significantdifference(P>0.05).3.The positive expression rate of MMP-2and IL-6invarious stage tissue of experimental group was higher than controlgroup(P<0.05).Expression of MMP-2increased with the progression ofinduced cancer, expression of IL-6in ICC lower thant-hat of atypicalhyperplasia stage, in turn was higher than liver inflammatory lesions, liverparenchyma hardening phase, with significant difference (P<0.05).4.Theexpression of MMP-2and IL-6correlated=each(P<0.05) other during theprogressio-n of ICC.5.There was no significant difference between differentdegree of differentiation ICC about MMP-2; the positive expression rate ofIL-6in high differentiation ICC was significantly lower than that of themiddleand low differentiation, respectively(P<0.05).Conclusion:1.ICC can be successfully induced by3`-Me-DBA in SDrats,this result can provides reliable method of animal model, which is simple,short cycle, low cost, strong repeatability,etc. But also provides a relevanttheoretical basis for improving the success rate of ICC model modeling infuture.2.MMP-2levels in bile have significant difference in the developmentof ICC, which can provide theory evidence for improving the early diagnosisof ICC.While, no significant difference of bile IL-6levels was detected in theICC development process, while suggested that bile IL-6levels can not beregarded as a specific diagnostic marker of the ICC.3.The expression of MMP-2and IL-6quantity in different phases of the ICC induced cancer havedifferences, this result can be as a obijective indicators which reflect the current stage of development of the ICC.4.According to the correlativeanalysis, MMP-2was positive correlated to IL-6in the process of ICCdevelopment, so ICC joint detection can be further improved diagnostic yieldmore accurate judgments of its development and prognosis.5.IL-6expressionlevels differ in varying degrees of differentiation ICC, while the ICC candetermine as a preliminary indicator of the degree of differentiation, helpsselect the appropriate therapeutic measures to improve the survival rate ofpatients with ICC. |
