Objective and background Insulin-like growth factor-1receptor(IGF-1R) and matrixmetalloproteinase-2(MMP-2) protein are overexpressioned in varied tumors andassociated with their biologic behavior.It has becoming a new hot spot of oncologytumor study recently.The insulin-like is one of the important factors for growthhormone and play an impertant role in every stages of body growth,it have the functionof regulating the growth,development of body,reproduction,immune function andmetabolites.MMP-2is an important member in matris metalloproteinases family,it candegrade the extracelluar matrix and was involved in physioklogical and pathologicalprocess,By induced from etracellular matrix metalloproteinase inducer,it can participatein the creation, the development, the metastasis and angiogenesis of the tumor.Toobserve the expression of IGF-1R and MMP-2protein in renal cell carcinoma and toinvestigate their cliniopathological significance.Thus,it is a good sense for earlydiagnosis and newly treatment for the renal cell carcinoma.Materials and Methods This speciments used to immunohistochemical study comefrom72cases of patients after nephrectomy,from october,2008to march,2010,whoadmitted in the Anhui Medical University for the treatment of renal cell carcinoma.In24hours after nephrectomy,these speciments were fixed,dehydrationed and saved by10%formalin.None of patients had received chemotherapy or radiotherapy beforeoperations.They include48renal clear cell carcinoma and other kinds of renalcarcinoma. There are37male and35female,the median age was62.4(range34-36).Inthe same time,33non-tumor renal tussues nearby tumors was proved to be nomal tissues by pathologica examination. Expression of IGF-1R and MMP-2protein in72specimens of renal cell carcinoma and non-tumor renal tussues were examined by usingpolymer detection system for immuno-histological staining and crosscompared withcliniopathological features of renal cell carcinoma. To analize the realationship of bothIGF-1R and MMP-2protein expression.Results High expression of IGF-1R and MMP-2protein were observed in69.4%(50/72)and72.2%(52/72) of renal cell carcinoma tissues respectively, significantly higher thanthose in non-tumor renal tussues39.3%(13/33,P<0.05) and27.3%(9/33,P<0.05).The high expression rate of IGF-1R and MMP-2were significantly associated with thetumor size, lymph node metastasis, Fuhrman classification and Robson staging(allP<0.05), which had no related to the tumor pathologic types, patient age, sex(allP>0.05); The expression of IGF-1R was positively correlativelated with the expressionof MMP-2in renal cell carcinoma (P<0.05).Conclusion High expression of IGF-1R and MMP-2protein in renal cell carcinomamay predict it can play an very important role in the occurrence and development inrenal cell carcinoma; The expression of IGF-1R and MMP-2protein in renal cellcarcinoma was realated to Fuhrman classification,the more expression of IGF-1R andMMP-2,the more Fuhrman classification; The expression of IGF-1R and MMP-2protein in renal cell carcinoma was realated to Robson staging,the more expression ofIGF-1R and MMP-2,the more Robson staging.High expression of IGF-1R and MMP-2protein are significantly correlated with the creation, the development and themetastasis of renal cell carcinoma, The higher expression of IGF-1R and MMP-2protein play an important role in the renal cell carcinoma, examination of expression ofIGF-1R and MMP-2may be useful for judgement of maligancy of renal cell carcinomaand reference of further therapy. |
