| Objective:This study try to offer reference to using cortex phellodendri slices rationally on clinic by comparing the difference of main chemical composition and drug action between cortex phellodendri slices and different particle size grains slices.Methods:we start the following research mainly contained chemical pharmacy and pharmacology research methods.①Determine the best decoction artwark of cortex phellodendri slices and different particle size grains slices by orthogonal design experiment.②Detect berberine in the decoction of cortex phellodendri slices and the decoction of different particle size grains slices by thin-layer chromatography.③Using high performance liquid chomatogram to determin the contentment of berberine in the decoction of cortex phellodendri slices and the decoction of different particle size grains slices,to erect the figer printing of chermical materials in the decoction of cortex phellodendri slices and the decoction of different particle size grains slices,to compare the different of the chemical composition in the decoction of cortex phellodendri slices and the decoction of different particle size grains slices.④Consideration effect of the absorbance from different dose of the decoction of cortex phellodendri slices and the decoction of different particle size grains slices by the inflam empirical method of mouse cavitas abdominalis capillary permeability accentuation by acetic acid.⑤Consideration effect of the swelling rate of ear from different dose of the decoction of cortex phellodendri slices and the decoction of different particle size grains slices by the inflam empirical method of mouse swelling of ear by dimethyl benzene.Results:①The best decotion artmethod optimization by orthogonal design experiment is the reagent being tested got8times water,drcocted3times and each time decoced1.5hours.②we can detection berberine hydrochloride by thin-layer chromatography.③The result of the content of berberine hydrochloride shows that the content of berberine hydrochloride in coarse powder was highest, and the content of berberine hydrochloride in optyma coarse powder was the least.④the chemical fingerprint of the comparison results show that in the decoction of cortex phellodendri slices and the decoction of different particle size grains slices similarity is more than0.9, indicating that the decoction of cortex phellodendri slices and the decoction of different particle size grains slices contained in the categories of the type of chemical composition has no obvious difference.(5)Effect of the decoction of different crushing degrees of Cortex Phellodendri on mouse peritoneal capillary permeability increase caused by acetic acid(HAc), and the results showed that compared with controls, cortex phellodendri slices3.64,1.82and0.91g/kg group, the most coarse powder1.82,0.91g/kg group, the coarse powder3.64and0.91g/kg group can significantly inhibit mouse capillary permeability incease caused by acetic acid (P<0.01, P<0.05), and the most coarse powder3.64g/kg group and the coarse powder1.82g/kg group compared with model, shows no obvious inhibition (P>0.05).⑥Result of the effect of the decoction of different crushing degrees of Cortex Phellodendri on mouse ear swell caused by Xylene show that compared with controls, cortex phellodendri slices1.82,0.91g/kg group, the most coarse powder3.64,1.82and0.91g/kg group and the coasrse powdel.82,0.91g/kg group can significantly inhibit mouse ear swell caused by xylene.(P<0.05), and cortex phellodendri slices3.64g/kg group and the coarse powder3.64g/kg group compared with controls, no significant inhibitory effect (P>0.05).Conclusions:①Effective component berberine hydrochloride dissolve is chang after smashing cortex phellodendri slices, and presents the trend that the decoction of the coarse powder> the decoction of the cortex phellodendri slices> the decoction of the most coarse powder, the decoction of the coarse powder content in the highest,but there is no significant difference.②There is no significant difference of the categories of chemical composition between the decoction of the cortex phellodendri slices and the decoction of the particle slices.③Cortex phellodendri yinpian and different pieces phellodendri yinpian degree is anti-inflammatory, but the same crushing degrees between different dose sample anti-inflammatory effects no significant differences. Tip:golden cypress yinpian and particles of slices of berberine, content and anti-inflammatory effects no significant differences. From the Angle of environmental protection evaluation, particle yinpian output less residue, suggest to use. |
