| ObjiectiveThe objiective of this project was to study the Monomer separation of the ethyl acetate part in Sedum aizoon L. On this fundation,we filtered out the hemostatic and anti-inflammatory active ingredients.This experiment was to provide theoretical basis for the clinical application of the effect of hemostatic and anti-inflammatory in Sedum aizoon L.Methods1.The ethyl acetate part were separated using chromatographic technique of silica gel,sephadex-20,HPLC and their structures were determined by the physical and chemical property and spectra of H-NMR,C-NMR and MS.2.The screening of hemostatic efficacy:capillary method of cultivating Sedum aizoon L.parts separation of ethyl acetate monomer effect on coagulation time (CT) of mice;Instrumental method of cultivating the separation of ethyl acetate monomer effect on prothrombin time (PT),activated partial thromboplastin time (APTT) and thrombin time (TT) of mice.3.The screening of anti-inflammatory efficacy:MTT assay for detection of cultivating Sedum aizoon L.parts separation of ethyl acetate monomer effect on RAW264.7mouse mononuclear phagocyte and the LPS stimulation the cell;Nitrate reduction test of cultivating Sedum aizoon L.parts separation of ethyl acetate monomer effect on LPS stimulation the cell release NO;ELISA method of cultivating Sedum aizoon L.parts separation of ethyl acetate monomer effect on LPS stimulation the cell release TNF-a, IL-iB and IL-6.Results1.The Sedum aizoon L.parts separation of ethyl acetate monomer were gallic acid,p-hydroxybenzoic acid,protocatechuic acid,vanillic acid,caffeic acid,5,7-two-color ketone,quercetin,kaempferol and luteolin.2.Yangxin grass ethyl acetate parts of the separation of phenolic acid compounds as gallic acid, vanillic acid and mignonette hemostatic effect, and gallic acid (including300g/mL), vanillic acid (including100g/mL) and mignonette (100, including30g/mL) optimal hemostatic effect. 3.Cultivating Sedum aizoon L.parts of ethyl acetate and separation of protocatechuic acid,vanillic acid,caffeic acid,quercetin and luteolin had anti-inflammatory efficacy.Conclusionl.The gallic acid,p-hydroxybenzoic acid,protocatechuic acid,vanillic acid and caffeic acid had hemostatic efficacy many activate many kinds of cruor factor in endogenous coagulation and extrinsic coagulation,and shorten CTã€APTTã€TT.2.The protocatechuic acid,vanillic acid,caffeic acid,quercetin and luteolin had anti-inflammatory efficacy may inhibite the generation of NOã€TNF-aã€IL-1Bã€IL-6pro-inflammatory cytokines. |
