Objective: To explore the treatmention mechanism of whole-body vibration onpostmenopausal osteoporosis, this study observed the effects of whole-body vibrationon serum sex hormone level, bone mineral density, and the protein expression ofPPAR γ and GSK-3β in hind limb of ovariectomized osteoporosis rats.Methods: Thirty-six three months old female SD rats were randomly dividedinto following three groups by body weight: sham-operation (Sham), ovariectomized(OVX), and ovariectomized vibration (Vibration). After ten weeks of OVX, theVibration group was treated with whole-body vertical vibration for15min at afrequency of90HZ two times a day. All vibration therapy was treated in the morningand the rats were sed in the same conditions. After7weeks of whole-body verticalvibration treatment, the whole-body bone mineral density, bone mineral content andbody composition was detected by dual energy X-ray absorptiometry. The changes ofserum and bone ALP levels as well as the serum estrogen (E2), progesterone (P),testosterone (T4), and triglycerides (TG) level were detected by721Photometer. Andthe protein expression of PPAR γ and P-GSK-3β was detected by western blot.Result:(1) The body weight in OVX group was significantly higher than that ofin Sham group. The body weight change in Vibration group was significantly lowerthan that of in OVX group.(2) There were no significant difference in serum E2, P4,and T between the Vibration group and OVX group, but significant difference wereseen in serum TG and TC. The bone ALP level in Vibration group was significantlyhigher than that of in OVX group.(4) The bone mineral density in distal femur andtibia were significantly higher than that of in OVX group while no significantdifference was seen in middleshaft.(5) The PPARγ protein expression in hind limb ofthe Vibration group was significantly lower that of in Vibration group while theprotein expression of P-GSK-3β was significantly higher.Conclusion: The whole-body vertical vibration could inhibit the increasing ofbody weight and fat mass.(2) The whole-body vertical vibration could decrease theserum level of TC and TG, but had no significant difference on serum level of sexhormones.(3) The whole-body vertical vibration could mitigate the bone lose of theOVX osteoporosis rats.(4) The whole-body vertical vibration could reduce theprotein expression of PPARγ in hind limb, but could increase the protein expressionof P-GSK-3β.
