ObjectiveTaking acute pharyngitis rats as animal models, evaluate the efficacy of acute pharyngitis with Yanyan Yin and preliminary study its mechanism.MethodsUsing15%ammonia water to make rat model of acute pharyngitis by pharyngeal spray method. All rats are divided into normal group, model group, shuanghuanglian group, Yanyan Yin high-dose, low-dose two groups. Yanyan Yin high-dose, low-dose groups are given7.2g/kg·d,3.6g/kg·d Yanyan Yin; Shuanghuanglian group is given shuanghuanglian oral liquid8ml/kg/d; Normal group and model group were given the same amount of normal saline,2times a day,for7days.Take the blood in the abdominal aorta for detection of white blood cell count and differential blood count.Centrifuge to obtain serum,and detect tumor necrosis factor-a (TNF-a), interleukin-1β (IL-1β), interleukin-6(IL-6) through Enzyme-linked immunosorbent assay(ELISA). Taking pharyngeal tissues, HE staining, observe the pathological change under light microscopy.Results1. Effect of Yanyan Yin on WBC and differential blood count of acute pharyngitis rats:Compared with the normal group,WBC count and differential blood count significantly increase in model group(P<0.05), that model is established successfully.Compared with the model group,WBC count and differential blood count significantly decline in Yanyan Yin high-dose, low-dose groups(P<0.05),that Yanyan Yin has the pharmacological effects of anti-acute pharyngitis.while High-dose group is better than shuanghuanglian group in reducing the neutrophil count, P<0.05.2. Effect of Yanyan Yin on pharyngeal tissue structure of acute pharyngitis rats:Under the light microscope, organizational structure of normal rats’ pharynx is clear, Mucosal epithelium is smooth, the lymphoid tissue and acinar scatter in the lamina propria. a small amount of small blood vessels in the submucosal,muscle fiber alignment rules.Organizations in the pharynx of model group rats is no clear. Mucosal epithelium cornification, fall off. Lamina propria and submucosal edema, hyperemia small blood vessels. A large number of inflammatory cells infiltration, a small amount into muscle tissue. Muscle fiber arrangement is irregular. Shuanghuanglian rats throat tissue mucosal epithelial less smooth, spikes significantly, with more mucus. Mucosal and submucosal inflammatory infiltration. Pharyngeal mucosa epithelium of Yanyan Yin low-dose group rats cornification, parts necrosis. Inflammatory cells gathered under the mucous membrane and mucous membrane,muscle fiber alignment rules." Small amount of residual mucus stay on pharyngeal mucosa epithelium of Yanyan Yin high-dose group rats. Small amount of inflammatory cell infiltrate in mucosa and submucosa.3. Effect of Yanyan Yin on inflammatory factors of acute pharyngitis rats:Compared with normal group, TNF-a,IL-1β,IL-6concentration significantly increase in model group. Compared with model group, high dose Yanyan Yin can obviously reduce TNF-a, IL-1β,IL-6concentrations (P<0.01),that Yanyan Yin has the pharmacological effects of reducing TNF-a, IL-1β, IL-6in serum of acute pharyngitis rats. And reducing TNF-a and IL-1β, Yanyan Yin high dose group is better than ShuangHuangLian group (P<0.05). Conclusion1. The Chinese traditional medicine can reduce the white blood cells of acute pharyngitis rats, that Yanyan Yin has the effect of anti-inflammatory.2. The Chinese traditional medicine Yanyan Yin make inflammatory cell infiltration in pharyngeal tissues reduce, inhibit the inflammatory reaction, and promote tissue recovery.3. Yanyan Yin can treat acute pharyngitis and the mechanism may be related to the inhibition of TNF-a, IL-1β, IL-6and other inflammatory factors secretion. |
