ObjectiveTo investigate the protective effect of Semen Astragali Complanali onkidney in a ratmodel of calcium oxalate nephrolithiasis.MethodsAdult male Sprague Dawley rats were given ethylene glycol andammonium chloride to induce urolithiasis for3weeks.Meanwhile, the ratsreceived the Semen Astragali Complanali extract at the doses of0.10g/mL、0.20g/mL、0.40g/mL, body weight, respectively. The establishment of normalcontrol group and pathologic group(n=10). Normal control group andpathologic group was given distilled water. After experiment3weeks,the24hurine concentration Ox、Ca2+ and HA of renal tissue were detected in each ofgroup rats,The calcium oxalate deposit in the kidney was observed bymicroscopy.Immunohistochemistry were used to assess the proteinexpression of CD44in rat renal tissue of every groups.Results1.Urine test index results:24h urine concentration Ox,Ca2+of stoneformation group and Extract from Semen Astragali Complanali interventiongroup were obviously higher than control group (P<0.01);Extract from SemenAstragali Complanali intervention group was lower than stone formation groupobviously (P>0.05).2. The content of hyaluronic acid in the renal tissues:HA of stoneformation group and Extract from Semen Astragali Complanali intervention group were obviously higher than control group (P<0.01);Extract from SemenAstragali Complanali intervention group was lower than stone formation groupobviously (P>0.05).3. Von Kossa’s dyeing and observe the pathological chang of kidney: VonKossa’s method clearly demonstrated the presence of pitchy calcium oxalatedeposits in the kidney of the stone formation group and Semen AstragaliComplanali intervention group.No deposits were detected in the controlgroup.The tubular dilatation in the kidney was fended on stone formationgroup and Semen Astragali Complanali intervention group.Quantitativeanalysis of the density of calcium deposits showed a significantly lowerdensity of the density of calcium deposits shower a significantly lower densityof deposits in the Semen Astragali Complanali intervention group compared tothe stone formation group.4.CD44protein immunohistochemical results:in rats normal controlgroup,only a weak CD44protein staining was noted.The expression wasenhanced in the stone group.Extract from Semen AstragaliComplanali-treated rats tended to have lower CD44protein expression thanthe stone group.Conclusions1.The extracts from Semen Astragali Complanali can block the formationof renal calcium oxalate stone in male rats potently.2.They may reduce the Renal injury and inhibit the expression of CD44and HA,thus inhibit calcium oxalate crystal deposition in rat kidney.
