| Objective:To investigate the expression level of Shh(Sonic hedgehog) and Gli1(glioma-associated oncogene1)in cervicitis, CIN, cervical carcinoma in variable and different stages and different pathological types.The study analyses the relationship between the expreesion level of Shh, Gli1and clinical pathological parametes.This can lay the foundation for Shh and Gli1may become the tumor markers of the judgement of the cervical cancer prognosis and become target genes of cervical cancer treatment.Methods:The total114cases of specimen were from vaginoscope biopsy or suegical operation in the partment of the gynaaecology of the second xiangya hospital of central south university during March2011to October2012.Among the14cases, there were24cases of cervicitis,22cases of CINI,23cases of CINⅡ-Ⅲ and45cervical cancer cases. The clinical specimen request patients with non history of cancer, and they were not given any radiation and chemotherapy.The clinical specimen organization obtained immediately bubble in10%formalin fixed in liquid, conventional paraffin embedding, sectioning, made four micros slice;First HE dyeing, to confirm the pathological diagnosisagain, then detected Shh and Gli1protein by using two steps immunohistochemistry;then observed the relationship between the expression change and the clinicopathological paramenters by using suitable statistics method.Results:1.The expression of Shh in cervical cancer, CIN, cervicitis and its clinical significance(1).In cervicitis, CINI, CINⅡ-Ⅲ to cervical cancer, the Shh positive rate are respectively16.7%,40.9%,65.2%,80.0%, The Shh positive expression rate was significantly different in the four groups(P<0.05).From cervicitis, CINI, CINⅡ-Ⅲ to cervical cancer, the Shh increased gradually. (2).The expression of Shh protein was not significantly related with patients’ages, clinical stages, presence of high risk HPV, pathological type,, presence of lymph node metastasis, and the degree of histologic (P>0.05).But the expression of Shh protein was significantly related with patients’clinical stages(P<0.05).2.The expression of Glil in cervical cancer, CIN, cervicitis and its clinical significance(1).In cervicitis, CINI, CINⅡ-Ⅲ and cervical cancer, the Gli1positive rate are respectively12.5%,45.5%,69.6%,86.7%, The Gli1positive expression rate was significantly different in the four groups(P<0.05).From nomal cervical, CINI, CINⅡ-Ⅲ to cervical cancer, the Gli1increased gradually.(2).The expression of Glil protein was not significantly related with patients’ages, presence of high risk HPV, pathological type, and the degree of histologic differentiation (P>0.05). But the expression of Gli1protein was significantly related with patients’clinical stages and presence of lymph node metastasis(P<0.05).3.Correlation analysis of Shh and Gli1protein expression in cervical cancerThe result showed that from cervicitis, CIN to cervical cancer, Shh and Gli1expression both showed a trend of increase gradually.Shh and Gli1expression showed significant positive correlation(R=0.458, P <0.05).Conclusions:1.Shh and Gli1increased gradually from cervicitis, CINI, CINⅡ-Ⅲ to cervical cancer. We can make a conclusion that Shh and Gli1play important role in the occurrence and development in cervical cancer.2.The expression of Shh protein was significantly related with patients’ clinical stages and the expression of Gli1protein was significantly related with patients’ clinical stages and presence of lymph node metastasis. We can make a conclusion that Shh and Gli1play important role in the invasion and metastasis in cervical cancer. |
