The Clinical Observation And Animal Experiments On Yi Shen Qing Li Granule’ Intervention Effects Of Inflammation Factors In The Dampness-heat Syndrome Of Chronic Kidney

Objective:The chronic glomerulonephritis is a immune-mediated inflammatory disease.Inflammatory cytokines is an important part of the immune system, and play an important role in the development of chronic glomerulonephritis.Through experimental research and clinical observation,this issue studied mechanism of immune regulation of Yi shen qing li Granule in damp-heat syndrome of chronic glomerulonephritis from the pro-inflammatory cytokines (TNF-a, IL-6, IL-8). To provide a solid theoretical foundation and a fuller scientific basis for Yi shen qing li Granule to treat damp-heat syndrome of chronic glomerulonephritis kidney.Methods:Through experimental research and clinical observation,this topic is in order to elaborate the solid theoretical foundation and fuller scientific basis for Yi shen qing li Granule to treat damp-heat syndrome of chronic glomerulonephritis kidney.(1) In the experimental study, We established the C-BSA glomerulonephritis rat model, the rats were randomly divided into five groups:the normal group, the positive control group,the model group, high-dose group of Yi shen qing li Granule and low-dose group of Yi shen qing li Granule. Testing24-hour urinary protein quantitative, renal function (serum creatinine, urea nitrogen), and detecting the level of IL-6in serum and the dynamic changes before and after treatment by double-antibody sandwich ELISA.(2) In the Clinical study, choosing30patients with chronic kidney of damp-heat syndrome which met the inclusion criteria syndrome type,then dividing them into two groups in accordance with the principle of randomized controlled, the control group was only given routine western medicine; the Test group was treated with routine western medicine+Yi shen qing li Granule. Detecting the level of TNF-a, IL-6,IL-8in serum and the dynamic changes before and after treatment by double-antibody sandwich ELISA.Results:(1) The experimental study reported that the24h urine protein, serum creatinine, IL-6of the Modeling rats were significantly higher than the control group (P<0.05, P<0.01).After treating by Yi shen qing li Granule,the serum urine protein, serum creatinine, IL-6levels of the rats in high-dose group and low-dose group of Yi shen qing li Granule were significantly lower than that the model group (P<0.05,P<0.01)The result suggested that Yi shen qing li Granule can significantly reduce the level of rat serum’s IL-6.(2) In the Clinical study after treatment,the24h urinary protein excretion, BUN, Scr, IL-6, IL-8and TNF-α level in both of the two groups were significantly improved than before treatment (P<0.05); Between the two groups, the24h urinary protein, Scr, IL-6, IL-8and TNF-a levels of the treat group decreased more significantly than the control group (P<0.05). The result Explained Yi shen qing li Granule can significantly improve patients’ symptoms, drop the urinary protein and reduce the level of TNF-α, IL-6, IL-8in serum.Conclusion:The above clinical and experimental results show that Yi shen qing li Granule play a significant role in the treatment of chronic glomerulonephritis kidney Damp-heat, it has a protective effect on renal function and its therapeutic mechanism may be related to inhibitiing expression of TNF-α, IL-6, IL-8which called proinflammatory cytokines, to blocking inflammatory response, inhibiting the release of inflammatory mediators.
