Objective: To investigate the effects of Erzhi Tiangui Granule on the kidney inpatients undergoing in vitro fertilization-embryo transfer (IVF-ET) cycle of transforminggrowth factor beta (lTGFβ1)expression levels in granulose cells, further explore kidney inChinese medicine improve the quality of oocytes and the mechanism of embryo quality.Methods:66routine IVF-ET treatment of kidney deficiency type of infertilitypatients were randomly divided into2groups, treatment group in33cases (Erzhi TianguiGranule joint designed the Gn particles observed group) and control group33casescombining designed the Gn group (placebo), adopt super ovulation long control scheme.Basic situation observed two groups of patients before treatment, kidney symptom scoreswere compared between before and after treatment, two groups of patients before treatmentendocrine follicle-stimulating hormone (FSH), estradiol (E2), the day of HCG serum E2level and the amount of a single oocyte E2level and designed the Gn clinical quality, wonthe number of oocytes, high quality oocytes rate, high quality embryo rate, fertilization rate,clinical pregnancy rate after treatment, application of real-time fluorescent quantitativepolymerase chain reaction (real-time policy polymerase chain reaction, real-time PCR)technology to detect follicle fluid TGF beta1expression in granulosa cells.Results:(1) Before treatment two groups patients’ basic situation, FSH and E2levelsbefore treatment, no statistical significance (P>0.05), comparable.(2) The treatment groupbefore and after treatment in patients with kidney deficiency symptoms have improvedsignificantly, the difference was statistically significant (P<0.05), while control groupkidney symptom improvement is not obvious, difference reached statistical significance(P>0.05).(3)The Gn dosage in treatment group was obviously lower than the control group,the difference was statistically significant (P<0.05).(4)The treatment group receivednumber of oocytes, oocyte quality rate, fertilization rate, high quality embryo rate andclinical pregnancy rates were significantly higher than the control group, the difference was statistically significant (P<0.05).(4)Treatment group follicle fluid granulosa cellsTGF beta1expression was higher than the control group, the difference was statisticallysignificant (P<0.05).(5)The follicular granulosa cells TGF beta1expression level andkidney symptom score after treatment showed a negative correlation (r=0.525, P<0.05),(6)Follicle granulosa cells TGF beta1expression was positively correlated withhigh-quality embryo rate (r=0.469, P<0.05), with the high quality oocytes though rate doesnot meet the positive correlation (r=0.192, P>0.05), but there are obvious positivecorrelation tendency.Conclusion: in IVF-ET treatment cycle, Erzhi Tiangui Granule can improve thesymptoms of patients with Deficiency of the kidney, reduce the dosage of the Gn, increasethe rate of oocytes with high quality, high quality embryo rate, clinical pregnancy rate canbe improved. Erzhi Tiangui Granule can improve the follicular particles cells TGF beta1expression level, thus improve the quality of the oocyte and embryo quality, this may beone of the mechanisms that Erzhi Tiangui Granule improve successful rate of IVF-ET.
