It mainly consists of interstitial dependentpluripotent basal-like cells, and can differentiate into epidermal or adnexal oflow-grade malignant tumor. Studies have shown that: the occurrence anddevelopment of the tumor is progressive multi-step, under the action of externaland internal factors combined result. Apoptosis in cancer cells, the activation ofoncogenes and inactivation of tumor suppressor genes, and the regulation ofcell proliferation split disorders is one of the important nature of the tumor. Theaims of this study is to observe by immunohistochemical methods and imageanalysis software analysis P16, CyclinD1, PTEN expression in BCC and therelationship between the P16,CyclinD1,PTEN and investigate the role andsignificance of BCC development. Provide the theoretical basis of the etiologyand pathogenesis of BCC.Methods: The expression of P16, CyclinD1and PTEN in40cases of BCCand15cases of normal skin tissues were detected by immunohistochemistryEnVision technique.5the horizons (×200) does not overlap were testedrandomly by OLYMPUS optical microscope BH-2,and captured images with aCCD digital camera. Collected images positive staining results of Collectedimages were semi-quantitative analyzed with the the Media Cybernetics professional image analysis software (image pro plu6.0, IPP), and evaluationof the average optical density (optical density mean) as positive staining. Fiveof the slices view of average optical density averaged then using SPSS17.0software as the average optical density of the slice, then the average opticaldensity of each group as the mean and standard deviation of the samplenormality test and the homogeneity of variance test. Proven skin basal cellcarcinoma and normal skin tissue protein average optical density follow anormal distribution, but the overall variance arrhythmia, then use twoindependent samples corrected t-test (t ’test) to analyze the differences of theBCC and normal skin tissuewhether in the expression of the three proteins withstatistical significance. Three kinds of protein in BCC pairwise relationshipwith the level of rank-related Spearman analysised.Results:1.The optical density mean of P16in BCC were lower than those incontrol group, the difference was statistically significant (P=0.00...<0.05).The optical density mean of CyclinD1in BCC were higher than those in controlgroup, the difference was statistically significant (P=0.00...<0.05). Theoptical density mean of PTEN in BCC were lower than those in control group,the difference was statistically significant (P=0.00...<0.05).2.The expressionof P16and CyclinD1were inversely correlated in BCC(r=-0.497), and theresult was statistically significant (P=0.00...<0.05);The expression of PTENand CyclinD1were inversely correlated in BCC (r=-0.546), and the result was statistically significant (P=0.00...<0.05);The expression of P16and PTENwere positively correlated in BCC(r=0.742), and the result was statisticallysignificant (P=0.00...<0.05).Conclusion:1. The optical density mean of P16in BCC were lower thanthose in control group,indicating that the gene has a correlation with thedevelopment of BCC.2. The optical density mean of CyclinD1in BCC issignificantly higher than normal skin tissue,its abnormal expression may beassociated with the occurrence and development of BCC,and may have as oneof the evaluation index of BCC.3. The optical density mean of PTEN in BCCwere lower than those in control group,showed that PTEN is closely related tothe base skin cancer proliferation.4. The expression of P16and CyclinD1wereinversely correlated in BCC, showed that they has relationship withproliferation of differentiation of BCC. Combined detection of P16andCyclinD1to determine the malignant degree and prognosis of BCC is helpful,could have important guiding significance and clinical value.5. CyclinD1andPTEN expression in BCC showed a negative correlation,and prompt them tothe BCC of mitotic cycle regulation exists in certain relationship. Jointdetection at the same time, both are likely to infer malignant degree andprognosis of BCC.6.
