Objective: Prescription of Rekeping injection is made up of Centipedaminima(L.) A. Br.et Ascher. and Bupleurum chinense DC,it’s antipyretic andanti-inflammatory effects are remarkable.As the most of TCM injections,compositions of Rekeping injection are complex,the active compositions arestill not clear.No matter home or abroad,research on Rekeping injection isscarce.The purpose of this study is to determine the effective part ofRekeping injection and improve the safety and effectiveness of that.Methods:First of all,optimize the extraction process of volatile ofCentipeda minima(VOCM) was determined by orthogonaldesign.Then,VOCM was separated by silica gel columnchromatography.Then, utilizing the swelling extent of rat toes induced byegg white and the swelling extent of mice ears caused by xylol,theanti-inflammatory effect of VOCM and each group after separation ofVOCM was observed,the abirritation of VOCM and their separatedcomponents was assessed by hot plate plan,Utilizing the raised extent ofrabbit temperature caused by endotoxin to observe the antipyretic effect ofVOCM and their separated components.Then,analyzed the components ofVOCM by GC-MS.Finally,determined the best ratio of the active components of VOCM and volatiles of Bupleurum chinense DC (VOBC) byanimal experiment.Results: The result of orthogonal design showed the best conditionswere as following which can get the most extraction ratio (0.35%,V/m):Soakthe Centipeda minima powder12h,the ratio of crude drug to liquid is9:1,theextraction time is6h.VOCM was separated to4parts by silica gel columnchromatography and marked as VOCM1,VOCM2,VOCM3andVOCM4.VOCM and each group after separation of VOCM didn’t showobvious inhibition on the raised temperature of rabbit caused byendotoxin.VOCM,VOCM2and VOCM3showed obvious inhibition onswelling rat toes induced by egg white,swelling mice ears caused by xyloland pain caused by hot plate plan.42,18,11,6and9compounds wereidentified from VOCM and VOCM1—VOCM4by GC-MS.The activeingredient of VOBC(VOBC2) have the most obvious antipyretic effect,thebest ratio of anti-inflammatory is8∶2(Centipeda minima: Bupleurumchinense DC).Conclusion:The best ratio of extraction process and prescription weredetermined and the main effective material basic of Rekeping injection hasbeen found by this experiment.The result of the experiment provides acertain basis for the development of new Rekeping injection.
