Objective: To study the chemical constituent from90%ethanol extraction and thebioactivities from compound of Dendrobium officinale and Dendrobium devonanum.Methods: The dried stems of D. officinale (2kg) and D. Devonanum (5kg) werelabeled as powder and EtOH (90%) were refluxed three times to get the EtOH extract.Successively extraction with EtOAc and n-BuOH to give three extraction parts: EtOAc,n-BuOH and water soluble portion. The EtOAc extract was purified by repeated columnchromatography with silica gel column, MCI, Sephadex LH-20(MeOH), and prepHPLC on Waters-Symmetry Prep C18column, to give compound. The structure of themonomer compounds were determined by mass spectrometry, nuclear magneticresonance date. The bibenzyls and its derivatives(compounds one to ten) from D.Officinale be used to do the experiments of neurite outgrowth-promoting activity. Someof compounds(compound5,8and9) from D. Devonanum be used to do the experimentsof neurite outgrowth-promoting activity and Acetylcholinesterase/Butyrylcholinesterase (AChE/BuChE) inhibitory activity.Results:17compounds has been isolated and identified from stem of D. Officinale, asfollows:4-(3,5-dimethoxyphenethyl) phenol (1),3-(4-hydroxyphenethyl)-5-methoxyphenol (2),5-(3-hydroxyphenethyl)-2-methoxyphenol (3),4-(4-hydroxyphenethyl)-2,6-dimethoxyphenol (4),4-(4-hydroxy-3-methoxyphenethyl)-2,6-dimethoxyphenol (5), gigantol (6),3-O-methylgigantol (7), dendrocandin T (8),dendrocandin U (9), dendrocandin B (10), erigeside п (11), clove aldehyde (12),2,6-dimethoxycyclohexa-2,5-diene-1,4-dione (13),(-)-syringaresinol (14),(+)-syringaresinol-4-O-β-D-monoglucoside(15), naringenin (16) and icariol A2-4-O-β-D-glucopyranoside (17).20compounds has been isolated and identified from stem of D.Devonanum, as follows: apigenin-7-O-glucoside (1), apigenin-7-O-rutin glycoside (2),erigeside п (3), citrusin C(4),6-(3,4-dihydroxy-5-(hydroxymethyl) tetrahydrofuran-2-yl) benzene-1,2,4-trio (5), icariol-A2-4-O-beta-D-glucopyranoside (6),(+)- syringaresinol-4-O-β-D-monoglucoside (7),1-(4’-hydroxy-3’-methoxypheny)propane-1,2,3-triol (8),1-(3’-hudroxy-4’-(hydroxymethyl) tetrahydrofuran-1’-yl)-5-methylbenzene-2,3,4-triol (9), gigantol (10), methyl-α-D-fructofuranoside(11),n-Butyl-α-D-fructofuranoside(12), methyl-β-D-fructopyranoside (13), n-propyl-β-D-fructopyranoside (14),3-methoxy-4-hydroxybenzaldehyde (15),4-(1’-hydroxypropyl)phenol (16), hydroxybenzotriazole formaldehyde (17),3-O-β-D-glucopyranoside-3,7-dimethyl-1,6-octadiene (18),3-O-β-D-glucopyranoside-3,7-dimethyl-1,7-oct-dien-6-one (19),3-O-β-D-glucopyranoside-3,6-dimethyl-1,6-heptadiene (20).compounds1,2,9and10from D. Officinale have a certain degree of neuriteoutgrowth-promoting activity and other compounds don’t have significant activity.compounds5,8and9from D. Devonanum don’t have activity in the experiments ofneurite outgrowth-promoting activity and Acetylcholinesterase/Butyryl cholinesterase(AChE/BuChE) inhibitory activity.Conclusion: Compound8and9are new compounds from D. Officinale. Compound8,9,18and20are new compounds from D. Devonanum. The bibenzyls and itsderivatives have some effect on PC12differentiation active, but the selected compoundsfrom D. Devonanum does not have this activity.
