component of Dendrobium,expecially of Dendrobium officinale,its polysaccharide content was specified that should not be less than 25%in Chinese pharmacopoeia.It has been proved that Dendrobium nobile polysaccharide has many pharmacological actions,although it is relatively less.Its alkali-soluble polysaccharide can tolerance swimming time in mice,raise blood sugar and triglyceride,reduce lactic acid and blood ammonia concentration,so decreased liver glycogen and muscle glycogen after swimming consumption.Modern research on Dendrobium candidum and Dendrobium nobile polysaccharide is limited to the water-soluble part,but tended to ignore the value of alkali soluble polysac-charide.According to the practical experience of taking fresh Dendrobium candidum,bubbling for 2 hours or repeatedly decocting still has certain viscosity.It can be spec-ulated that the Dendrobium may contain many other mucilaginous substance like alkali-soluble polysaccharide or acid-soluble polysaccharide.Alkali-soluble polysaccharide with sulfate radical has strong activity of antiviral,antit-umor or antioxidant,however,many alkali-soluble polysaccharide in the absence of sulfate radical.Modification and biological activity of polysa-ccharide sulfate structure study will be the future research focus for a long time.Objective:1.To establish a method for characteristic spectrum of lkali-soluble polysaccharide and water-soluble polysaccharide from seven species of Dendrobium by HPLC.2、To hint the importance of alkali-soluble polysaccharide by determining the content of the preliminary study of alkali-soluble and water-soluble polysaccharide from seven species of Dendrobium.3、To determine the biological activity of alkali-soluble and water-soluble polysaccharide from Dendrobium officinale Kimura et Migo and Dendrobium nobile Lindl.Methods:1、PMP-HPLC fingerprint of alkali-soluble polysaccharide and water-soluble polysaccharide from seven species of Dendrobium.2、Determination of the content of alkali-soluble and water-soluble polysaccharides of 7 species of Dendrobium by HPLC.3、Biological activity of the preliminary study of the alkali-soluble polysaccharide of Dendrobium officinale with wild simulated,the culture of Dendrobium officinale and Dendrobium nobile.,including determination of value-added rate of polysaccharides on Hela cells by WST and antioxidant tests in vitro.Results:1、The results shows that different Dendrobium alkali-soluble polysac-charides and water-soluble components has different monosaccharide content and content ratio.Seven species of Dendrobium’ s ratio of D-mannose and D-glucose:Dendrobium officinale are 3.026%-7.181;Dendrobium nobile are 0.163-2.098;Dendrobium moniliforme are 3.297-4.734;Dendrobium devonianum Paxt.are 2.628-3.634;Dendrobium aphyllum are 1.729-2.064;Dendrobium crepidatum are 0.808-1.089;Dendrobium chrysanthum are 1.085-1.141.But it’ s still hard to differentiate Dendrobium officinale Kimura et Migo by pharmacopoeia method.The monosaccharide composition of alkali-soluble polysaccharides more complex,which is mainly D-mannose,D-galacturonic acid,D-glucose,D-galactose,D-Xylose.There are many differences among the alkali-soluble polysaccharides of Dendrobium officinale Kimura et Migo,Dendrobium devonianum Paxt.and Dendrobium moniliforme(Linn.)Sw..In this way,Dendrobium officinale Kimura et Migo can be able to discriminated.2、The determination results are as follows:The content of water-soluble and alkali-soluble polysaccharide of Dendrobium officinale are 21.94-34.72%and 5.02-21.80%respectively.But 2.27-21.63%and 6.09-24.56%respectively for Dendrobium nobile,24.2-28.79%and 15.5-17.10%respectively for Dendrobium moniliforme,12.9-18.82 and 5.14-5.42 respectively for Dendrobium devonianum Paxt.,22.3-25.57%and 3.20-3.79 respectively for Dendrobium aphyllum,40.5-43.66%and 11.1-13.63%respectively for Dendrobium crepidatum,33.2-36.03 and 11.5-13.61 respectively for Dendrobium chrysanthum.It can be seen that alkali-soluble content is just3-0.9 times less than the water-soluble content in Dendrobium,except Dendrobium nobile Lindl.Higer.So the alkali-soluble polysaccharide of Dendrobium can not be ignored.It has important significance for the determination of total polysaccharides content.3、The reslut shows that both alkali-soluble and water-soluble polysaccharide could inhibit the activity of Hela cells.More over,the effect on cleaning free radical(· OH)and H202 of alkali-soluble polysaccharide is stronger than water-soluble part,but DPPH free radical scavenging activity just contray.These figures show that alkali-soluble polysaccharide of Dendrobium has high development and used value.Conclusion:the structure of alkali-soluble polysaccharide is likely different from water-soluble polysaccharide in Dendrobium biological activity as well.It is necessary to explored in depth.Polysaccharide is one of the most important effective components in Dendrobium officinale.It can be found from experimental results that the content of water-soluble polysaccharide of Dendrobium officinale which is not the most abundant in all Dendrobium species,but has higher nutritional and medicinal value,Dendrobium nobile as well.However,the content of alkali-soluble polysaccharide of the above two species are more than water-soluble parts.The relationship between medicinal value of the above two species and their alkali-soluble polysaccharide yet to be proved. |
