| Objective:To observe the effects of Shenfu Injection and the extract of Hongshen and Fuzi to rat and myocardial cell with heart failure and study the mechanism of Shenfu injection for heart failure.Methods:The study includes animal part and cell part. In the animal part, the rats are intubated in the left ventricular. After injured by Propafenone injection which is regard as heart failure model, the rats are treated with distilled water, Shenmai injection (6.67mL/kg), extract of Fuzi (3.33mL/kg), extract of Hongshen (3.33mL/kg) and Shenfu injection (6.67ã€3.33ã€1.67mL/kg) by femoral vein. Observe and record the change of HR,+dp/dtmax and-dp/dtmax following administration of5,10,20,30and60min. And detect the content and activity of Ang-â… , Ang-â…¡, ANP, ALD, ET and TNF-a in plasma of the rats. In the cell parts, the myocardial cells of baby rats are used. The myocardial cells are gotten by trypsin digestion and purified by different velocity of adherence. After injured by0.8%pentobarbital sodium, the myocardial cells are administrated with3different concentrations of Shenfu Injection (5ã€10ã€20μL/mL), extract of Fuzi (6g/L), extract of Hongshen (3g/L) and different compatibility between the two extracts (1:2,2:1,1:1). Then mensurate the vitality of the cells, the activity of the Ca2+-Mg2+-ATP enzyme, Na+-K+-ATP enzyme and Ca2+-ATP enzyme and the content of Na+, Mg2+, K+, Ca2+in the cell1h later.Results:In the animal part, the values of HR and+dp/dtmax are decreased,-dp/dtmax is increased, and the content of Ang-â… , Ang-â…¡, ANP, ALD, ET and TNF-a are increased after injured by Propafenone injection. After administrated with different testing drug, the values of HR and+dp/dtmax are increased,-dp/dtmax is desreased, and the content of Ang-â… , Ang-â…¡, ANP, ALD, ET and TNF-a are decreased with varying degress. In the cell part, the strength of the myocardial cell beating is decreased or disappeared, the activity of the Ca2+-ATP enzyme and Ca2+-Mg2+-ATP enzyme and the content of Na+, Mg2+are decreased, the activity of Na+-K+-ATP enzyme and the content of K+, Ca2+are increased after injured by pentobarbital sodium. The Shenfu Injection can increase the strength of the model myocardial cell beating, and enhance the activity of Ca2+-ATP enzyme and the Ca2+-Mg2+-ATP enzyme, inhibit the the activity of Na+-K+-ATP enzyme, increase the content of Na+, Mg2+and decrease the content of K+, Ca2+in the myocardial cell. To the normal myocardial cell, the testing drugs have the similar effects to model myocardial cell, but the effects are weaker.Conclusions:The Shenfu injection, extract of Fuzi and extract of Hongshen all have the effects to improve heart failure status and protect the heart. For rats, these drugs take effect by regulating the heart failure related cytokine and neurohormonal factor, which the extract of Fuzi and the high dosage of Shenfu injection better. For the myocardial cell, these drugs take effect by increasing the vitality of cells and regulating the internal environment of cells, which the compatibility (2:1) of Fuzi and Hongshen and the high dosage of Shenfu injection better. |
