Object:The Research intends to analyze the therapeutic efficacy of SodiumHyaluronate on xerophthalmia pattern of rabbits, which reveals its function ofmucoprotein imitation and way of reconstructing physiological tear mechanism.Materials and Method:48healthy New Zealand Rabbits, ranging from2.5kg to3kg, divided into4groups, which means a dozen rabbits every group. Right eyes were chosen to do theexperiment. Group A was the group treated with0.1%Sodium Hyaluronate EyeDrops, Group B was the group treated with0.3%Sodium Hyaluronate Eye Drops,Group C was the control group treated with Physiological Saline, while Group D wasthe blank control group. Rabbits in all four groups were subcutaneously injected withScopolamine Hydrobromide in the cervical part,1.0mg/kg, at8:00,11:00,14:00,18:00every day, continuing for35days. After injected for7days, Group A started tobe treated with0.1%Sodium Hyaluronate Eye Drops, Group B with0.3%SodiumHyaluronate Eye Drops, while Group C with Physiological Saline, continuing for28days. Tear secretion experiments, breakup time for tear film (BUT),keratoconjunctivitis lissamine green dying, cornea fluorescein dying, crystallizationexperiments for tear ferns, conjunctival mark cytolgical examinations were done atthe7th,14th,21st,28th and35th day. Keratoconjunctivitis lissamine green dying,cornea fluorescein dying, crystallization experiments for tear ferns were done forGroup A and Group B5minutes,30minutes,60minutes and90minutes after theywere treated with Sodium Hyaluronate Eye Drops.6rabbits from each group werekilled by overdosing Ketamine in random to get the cornea and conjunctiva forexperiments at the21st day and35th day. The expression of mucoprotein MUC5ACand MUC1, and extracellular matrix accepter were observed underimmunofluorescence technique. The rebuilding and renovation of corneal epitheliumwere observed under scanning electron microscope. Results:1.7days after injected with Scopolamine Hydrobromide, cornea fluoresceindying for the rabbits showed positive. Tear secretion volume was less than11.0±1.59mm.2. Comparing with the control groups, keratoconjunctivitis lissamine greendying, cornea fluorescein dying and tear ferns scores for Group A were notstatistically significant90minutes after the first treatment. Comparing with thecontrol groups, keratoconjunctivitis lissamine green dying, cornea fluorescein dyingscores for Group B were statistically significant (P<0.05) after30minutes after thetreatment, while tear ferns score was not statistically significant90minutes after.3. Tear secretion volume of the4groups during the experiment was notstatistically different.4. Comparing with the control groups, keratoconjunctivitis lissamine greendying score for Group A decreased and showed a statistically significant difference(P<0.002) at the28th day, but no statistically significant difference at the35th day.Group B showed a statistically significant difference (P<0.001) at the28th day andlasted to the end of the experiment. Cornea fluorescein dying score decreased andshowed a statistically significant difference (PA<0.001,PB<0.001) at the28th dayand lasted to the end of the experiment.5. Comparing with the control groups, the score of crystallization experimentsfor tear ferns for Group A decreased and showed a statistically significant difference(P<0.02) at the14th day, and lasted to the28th day(P<0.01). Group B showed astatistically significant difference (P<0.04) at the14th day and lasted to the end of theexperiment.6. Comparing with the control groups, the breakup time for tear film (BUT) ofGroup A and Group B increased, showed a statistically significant difference(PA<0.006,PB<0.005) at the28th day and lasted to the end of the experiment.7. Comparing with the control groups, conjunctival mark cytology showed astatistically significant difference(PA<0.006,PB<0.005) at the35th day.8. Comparing with the control groups, expression of Muc5Ac、Muc1for GroupA and Group B are significantly higher than that of Group C and Group, while expression of CD44for Group A and Group B are lower than that of Group C andGroup D.Conclusion:Sodium Hyaluronate Eye Drops are not only able to imitate tear mucoprotein’sthe function of stabilizing tear film, holding water, keeping wet and lubricating, butalso able to facilitate the proliferation and renovation of corneal epithelial cells,relieve corneal epithelial inflammation, and restore the damage of ocular surface, soas to treat the xerophthalmia effectively.
