Our aims of this study are to damaging of learning and memory ability by thedisease and the intervention effect of traditional Chinese medicine BXD under the behaviorof model, studying the effect of disease and traditional Chinese medicine BXD capsule tothe change of their learning and memory ability of the premenstrual mood dysphoricdisorder (PMDD) gan-qi invasion rat model.To probe the pathogenesis of PMDD with liver-qi invasion syndrome and research theintervention mechanism Baixiangdan Capsule for the disease, studying the effect of diseaseand traditional Chinese medicine Baixiangdan Capsule to the change of microscopicindicators in hippocampus NMDAR1receptor.Methods:⑴Model Preparation and Evaluation: Prepared the gan-qi invasion rat modelwith emotional-stimulation dominated multi-factor phased stimulation, and then intervenedthe model with Baixiangdan Capsule or Fluoxetine, and evaluate all the group rats by openfield test.⑵B ehavior Evaluation of learning and memory in rats: We detect learning andmemory abilities of the premenstrual mood dysphoric disorder (PMDD) gan-qi invasion ratmodel in the macroscopic behavior by Y-maze test, new-object recognition and MWM experimental.⑶Detect micro-indicators:Then we adopt the immune fluorescence taggingtechnology and Western blot technology to detect the protein distribution and expressionlevels of NMDAR1in CA1, CA3of Hippocampus in the normal group rats, model grouprats, Baixiangdan treatment group rats and Fluoxetine treatment group rats, and calculatethe mean fluorescence intensity in all group rats.Results:⑴Model Preparation and Evaluation: Compared with the normal group rats, thetotal distance moved incrsesed significantly (p<0.05), number of crosses into the centralarea and duration of movement in the central area decrsesed significantly(p<0.01,p<0.01)in model group rats. On the indicator the total distance moved decrsesed significantly(p<0.05), number of crosses into the central area and duration of movement in the centralarea Baixiangdan treatment group incrsesed significantly(p<0.05,p<0.05)compared to themodel group, On the indicator the total distance moved the Fluoxetine treatment groupdecrsesed significantly (P<0.05) compared to normal group.⑵B ehavior Evaluation of learning and memory in rats: Compared with the normalgroup rats, DI decrsesed significantly(p<0.05)in model group rats, DI incrsesedsignificantly(p<0.05)in Baixiangdan treatment group rats. In Morris water maze, placenavigation training, the model group rats escape latency obviously delayed in2,4days.Compared with the model group rats, Baixiangdan treatment group rats escape latencyobviously reduced(p<0.05, p<0.05). spatial probe test, compared with the normal grouprats, the model group, the escape latency obviously delayed(p<0.05), the number ofthrough platforms significantly reduced(p<0.05). Compared with the model group rats,Baixiangdan treatment group of the number of through platforms significantlyincrsesed(p<0.05).⑶Results of micro-indicators showed that: immune fluorescence test demonstratethat model group of the positive Hippocampus CA1, CA3pyramidal cell layerarrangement were sparse and disorder, cell gap large, compared with the control group.While Baixiangdan treatment group rats and Fluoxetine treatment group rats of the positiveHippocampus CA1, CA3pyramidal cell layer arranged in order, compared to model group. Compared with the normal group, Western blot test show that the protein expresstion levelsof NMDAR1have a lower(p<0.0001, p<0.001)in Hippocampus of the model group ratsand Fluoxetine treatment group rats. Compared with the model group rats and Fluoxetinetreatment group, the protein expression levels of NMDAR1in Baixiangdan treatmentgroup have a significant increase in the hippocampal (P<0.001).Conclusion:⑴Anxiety state of the premenstrual mood dysphoric disorder (PMDD)gan-qi invasion rat model can damage learning and memory abilities, While defects inlearning and memory abilities of the model rat can be corrected by Baixiangdan capsules,and the treatment effect may be superior to fluoxetine.⑵The low expressions of the NMDAR1Recepter subunit protein in Hippocampusand the positive Hippocampus CA1, CA3pyramidal cell layer reduced may have closerelationship with the premenstrual mood dysphoric disorder (PMDD) gan-qi invasion ratmodel. and Baixiangdan Capsule can correct the low expression of NMDAR1Receptersubunit in Hippocampus and the reducing of NMDAR1protein activaty, that probably isone of the centre pathogenesis of Baixiangdan capsule treating PMS with GanQi invasionsyndrome. |
