| Objective:To investigate the changes between protein location and expression levels ofγ-Aminobutyric Acid B1Receptor and γ-Aminobutyric Acid B2Receptor in Frontalcortex,Parietal cortex,Hippocampus and Hypothalamus and the relevant proteinexpression levels of extracellular signal regulated protein kinase(ERK) signal transductionpathway in Hippocampus of Premenstrual Syndrome Model Rats with liver-Qi invasionsyndrome, which gives a preliminary discussion on the pathogenesis of PMS with liver-qiinvasion syndrome,as well as researches the intervention mechanism Baixiangdan Capsulefor the disease.Methods: After open field test, macro-behavior observation and vaginal smearexamination, we choose the Wistar rats of health and female into test, then build PMSmodel rats with liver-Qi invasion syndrome by electric stimulation,and evaluate all thegroup rats by open field test and macro-behavior observation. Then we adopt the immunefluorescence double-tagging technology to detect the protein distribution and expressionlevels of GABABR1and GABABR2in Frontal cortex,Parietal cortex,CA1and CA3ofHippocampus and Hypothalamus in the normal group rats,model group rats, Baixiangdan treatment group rats and Baclofen treatment group rats,and calculate the mean fluorescenceintensity in all group rats. And we use Western blot technology to detect the proteinexpression levels of ERK1/2,P-ERK1/2in Hippocampus in all group rats,and calculate theP-ERK1/2percentage of ERK1/2.Results: Compared with the normal group rats, the crossing score,rearing score and totalscore of open field test all increased obviously(P<0.01)in model group rats.Meanwhile, therats in model group look like nervous, unstable and sensitive to surroundingstimulation;On the other hand,compared with the model group rats, the open field scoresall decreased remarkably(P<0.05or P<0.01)in the two treatment group rats. Comparedwith the normal group rats, immune fluorescence double-tagging test demonstrate that theprotein expression levels of GABABR1and GABABR2in model group rats decreasedremarkably(P<0.01) in Frontal cortex,Parietal cortex,CA1and CA3of Hippocampus andHypothalamus. Compared with the model group rats, the protein expression levels ofGABABR1and GABABR2in Baixiangdan treatment group and Baclofen treatment grouphave a significant increase in the every cortex(P<0.01or P<0.05), but there was nostatistical differences in the two treatment groups(P>0.05). Compared with the normalgroup, Western blot test show that the protein expresstion levels of P-ERK1/2have a lower(P<0.01)in Hippocampus of the model group rats; Compared with the model group rats,theprotein expresstion levels of P-ERK1/2increases(P<0.01) in Hippocampus of the twotreatment group rats,and there was a obvious increase between the Baixiangdan andBaclofen treatment groups(P<0.01).Conclusion: PMS Model Rats with liver-qi invasion syndrome can be preparedsuccessfully by using electric stimulation; The low expressions of the GABABReceptersubunit protein in Frontal cortex,Parietal cortex,CA1and CA3of Hippocampus andHypothalamus and P-ERK1/2protein in Hippocampus may have close relationship withPMS liver-qi invasion syndrome;and Baixiangdan Capsule can correct the low expressionof the protein in above brain regions,that probably is one of the centre pathogenesis ofBaixiangdan capsule treating PMS with liver-qi invasion syndrome. |
