| Objective:To investigate the association on the polymorphisms of endothelin family gene and migraine in the Chongqing Han ethnic group.Methods:We have performed an association study in304patients with migraine and304healthy controls. im-LDR DNA sequencing was used to detect7single nucleotide polymorphisms (SNPs) in4genes associated with endogenous endothelins synthesis. The analysis of their allele frequency, genotype and haplotype construction were performed.Results:Of the304migraineurs, the most frequently migraine symptoms were moderate or severe headache (99.0%), aggravated by routine activity(88.5%), phonophobia (81.6%),nausea (72.4%)and pulsatile pain (57.6%).There were no significant differences of headache characteristics between male migraineurs and female migraineurs.The frequencies of the endothelin family gene genotypes and allelic variants did not differ significantly between migraine patients and controls, and were unrelated with the presence of aura. However, the haplotypes analysis shows that a haplotypes of EDNRA gene (A-T-C-T, A-C-C-C) were associated positively with migraine with aura(p<0.05).Conclusion:In this sample,two haplotypes of EDNRA gene were significantly associated with migraine with aura, which suggest it may be a influencing factor of migraine in South Chinese Han. To confirm this finding, studies on even larger samples are required. |
