Object: To study an ideal preparation of Traditional Chinese Medicine for treatingHyperlipidemia. Methods: Single factor experiments and orthogonal experiment were usedto optimize the conditions of extraction process and refining process of Panaxnotoginseng,the the rate of notoginsenoside R1, ginsenoside Rg1and ginsenoside Rb1as indicators. Bysingle factor experiments and orthogonal experiment, the rate of epiberberine, coptisine,palmatine and berberine as indicators, the conditions of extraction process and refiningprocess of Coptis chinensis Franch were determined. The preparation process was studiedby screening of optimum ratio of panaxnotoginseng and Coptis chinensis Franch. Thequality standard was developed with notoginsenoside R1, ginsenoside Rg1, ginsenoside Rb1,epiberberine, coptisine, palmatine and berberine in the preparation which determined byHPLC. Metabolism of medicine was discussed by serum pharmacokinetics experiments.Results: The optimal extraction technology of Radix Notoginseng was as follows:extracted2times with50%ethanol(10times and8timesvolume). The optiumu refiningprocess wasas follows: the decompression recovery was used until the extraction containing0.3g crude medicines per milliliter, through HPD-100type macroporous resin column,eluted with water to the outflow of fluid nearly colorless, then eluted with80%ethanolthree times of the resin volume. The optimal extraction technology of Coptis chinensisFranch was as follows: extracted2times with70%ethanol (10times and8times volume).The optiumu refining process was salting-out and ion-exchange resin method. The optiumutablet moulding technics were as follows: roll granulation, the ratio ofactive component ofmodifying lipid, microcrystalline cellulose, magnesium stearate, talcum powder and sodiumcarboxymethyl starchwas65∶35∶1∶2∶3. The preparation has fine reproducibility andstability. The preparations reduce total cholesterol(TC), low density lipoprotein-cholesterol(LDL-C) and very low density lipoprotein-cholesterol (VLDL-C); simultaneously increasehigh-density lipoprotein cholesterol(HDL-C) and reduce triglyceride(TG). Conclusion:The preparation process is feasible and reasonable. The quality standard is suitable for thequantitative analysis. This preparations is an optimum preparation which is used to treatHyperlipidemia. The studies of the pharmacodynamics indicated that it has obviousTreating Hyperlipidemia effect. |
