Paeonia veitchii Lynch, belongs to the genus Paeonia, species Ranunculaceae,is also named Radix Paeoniae Rubra, Paeonia lactiflora Pall. It is as a commonsource of the herbal medicine Radix Paeoniae Rubra used in Chinese Pharmacopoeia2010. The root of the plant has been used as a traditional medicine for thousand yearsfor the treatment of promoting blood circulation to remove blood stasis. It’s slightlybitter taste, and slightly cold, medicine to the liver.The Paeonia veitchii is clinically valuable for applications in blood disease likeblood stagnation amenorrhea, herpes simplex, postpartum extravasated blood, dryblood and nosebleed nasally, warm poison and typhus. Modern pharmacologicalstudies have shown that the total glycosides of paeony have anticoagulation,anti-thrombosis, anti-endotoxin, anti-tumor, anti-depressionand effects and amelioratelearning and memory ability etc. The paeoniflorin has the function of neuroprotective,anti-diabetics, immuoregulation, anti-arthritis, and the protection of liver injury andliver fibrose etc.For further study of the medicinal value of the genus Paeonia, this paper firstoutlined the chemical and anti-tumor researches of Paeonia veitchii Lynch domesticand abroad. We also classified the constituents in this plant based on structuralfeatures. Secondly, systematical chromatorgraphic methods (D101macroporous resin,silica gel, octadecylsilyl column chromatography and the semi-preparing liquidchromatography) were carried out on the n-butyl alcohol extraction of the ethanolextract, for separation of the monoterpene glycosides compounds. Fifteen compoundswere isolated from the extract. Six of them were identified on the basis of spectralanalysis and physicochemical evidences. They are paeoniflorin (PN-02/PN-03),4-O-methyloxypaeoniflorin (PN-05),4-O-methylpaeoniflorin (PN-06),4-O-ethylpaeoniflorin (PN-08), paeonidanin (PN-12),9-ethyl-neo-paeoniaflorin A(PN-15). Compounds PN-05、PN-06and PN-12were isolated from this plant for thefirst time. Finally, studies of in vitro antitumor activity of PN-02~PN-15were investigated.The result showed that PN-02/PN-03, PN-06and mixture PN-07were active tohuman oral epidermoid carcinoma cell line KB, and PN-02/PN-03, PN-06, mixturePN-07and PN-15were active to human lung cancer cell line A549. It is worthy offurther study on the structure-activity relationship with compound PN-02, PN-03,PN-06, mixture PN-07and PN-15.We had made a systematically separation and purification on themonoterpene glycosides compounds of Paeonia veitchii Lynch for the firsttime. Preliminary results show that, paeoniflorin,4-O-methyloxypaeoniflorin,9-ethyl-neo-paeoniaflorin A and mixture PN-07were active to cancer cells. Itcan be useful for further study of the characteristic composition of Paeoniaveitchii Lynch.
