For the study whichelaborate chemical constitution of Apocynumm venetum L. and anti-platelet agglutinatingactivity and chemical constitution of Aucklandia lappa Decne. and antibacterial action ofStreptococcus mutans and their antioxidant activity, in order to supply some datainformation to complete substance basis of the two crude drugs. Methods:1. To preparesolvent distribution components from95%extracts of the two drugs by Petroleum, ethylacetate and n-butanol, macroporous absorbing resin components which using differentseparation material (such as silicon gel, macroporous absorbing resin and polyamide)different solvent system (Petroleum-ethyl acetate, chloroform-methanol, differentconcentrations of ethanol) by further prepare. These fractions were systemic analyzedadopting HPLC-PDAã€NMR.2. To determinate the contents of total flavonoids and totaltannin.3. To study antioxidant activity of two drugs’ different components throughdeterminate scavenging capacity of DPPH· radicals, Hydroxyl free radicals, Oxygen freeradicals and reducing capacity.4. Nephelometery was applied to evaluate anti-plateletagglutinating activity of multi-Components of Apocynumm venetum L.; Constant brothdilution method was used to measure MIC and MBC values antibacterial action ofStreptococcus mutans. of multi-components of Aucklandia lappa Decne. Results:1.29components were separated from Apocynumm venetum L.;16components were separatedfrom Aucklandia lappa Decne., costunolide (5mg) and dehydrocostus lactone(7.4mg)the main antibacterial component were separated from ethyl acetate comonennt ofAucklandia lappa Decne. by preparative chromatography.2. The contents of totalflavonoids and total tannin were high in ethyl acetate component and n-butanolcomponent of Apocynumm venetum L. were33.1763%and1.346%.3. Ethyl acetatecomponent had better r antioxidant activity in Apocynumm venetum L. and Aucklandia lappa Decne. L-â…¢, L-â…£ and M-â…¢, M-â…£ had better scavenging capacity of DPPH radicals; L-â…¡was better than Vc on scavenging Hydroxyl free radicals, M-â…¢ was betterthan other components; L-â…¡, L-â…¢ and M-â…¢was better on scavenging Oxygen freeradicals; L-â…¢ and M-â…¡had better reducing capacity.4. Ethyl acetate component (L-â…¢)and L-â…¢-4have better anti-platelet agglutinating activity than other components, weconcluded that isoquercitrin, hyperoside, lutin and other flavonoids have anti-plateletagglutinating activity to some extent; Petroleum component (M-â…¡), M-â…¢-2, costunolideand dehydrocostus lactone have better antibacterial action of Streptococcus mutans.Conclusion: systematic separation components were characterized by HPLC analysismethod, in vitro screening active components by anti-platelet agglutinating andantibacterial study, in order to associate two drugs’ chemical constitutes with activecomponents, and supplied data information for construction of Multi-Components Libraryof Uygur Medicine Aucklandia lappa Decne. and Apocynumm venetum L. |
