Effect Of Muxiang Granules On UC And Its Mechanism

A large number of evidence-based clinical studies have shown that intestinal mucosal barrier integrity was a core factor in the treatment and prevention of Ulcerative colitis.Intestinal mucosal barrier dysfunction or increased intestinal mucosal permeability can trigger or accelerate the occurrence and severity of immune-mediated colitis.It was an effective way to control ulcerative colitis clinically by repairing the intestinal mucosal barrier and controlling intestinal mucosal inflammation.Objectives:Search Chinese herbal medicine to repair the intestinal mucosal barrier.Methods:This research was based on Traditional Chinese Medicine and modern basic research.On the one hand,based on the clinical data in the real world,the concept and method of data mining was used to construct the TCM research based on ulcerative colitis in the real world.On the other hand,from the classical literature of traditional Chinese medicine,network pharmacology and scientific pharmacodynamic experiments,explore the mechanism of the effect of Radix Aucklandiae in the treatment of ulcerative colitis.Results:According to the results of real world studies,Aucklandia lappa Decne.was one of the core prescription oral drugs of Chinese herbal medicine in the remission period of ulcerative colitis.According to statistics of large-scale comprehensive materia medica literatures,Aucklandia lappa Decne.’s intervening was still in the gastrointestinal disease,the main diseases were dysentery,abdominal pain,and diarrea.Through reviewing the latest research results of Aucklandia lappa Decne in recent years,it was found that the active ingredients of Aucklandia lappa Decne were concentrated in the volatile oil,costunolide and dehydrocostus lactone were up to 50%volatile oil ingredients,and the main active ingredients had good anti-inflammatory and anti-tumor effects.These active ingredients associated with UC related inflammatory pathways(NF-Kb,TNF,PGE2,IFN),and intestinal mucosal barrier related protein(MUC2,Tight junction protein ZO-1).Basic experimental researches were also confirmed that Aucklandia lappa Decne and dehydrocostus lactone alleviated intestinal damages caused by DSS,reduced the colonic inflammation histological assessment,decreased UC related inflammatory cytokines(TNF-α,IL-6,IL-17,IL-23),repaired the key intestinal mucosal barrier protein(MUC2),inhibited the downstream pathway(a-defensin).Aucklandia lappa Decne and dehydrocostus lactone possesse the potential of anti-inflammatory effect to UC.The protective mechanism of Aucklandia lappa Decne and dehydrocostus lactone may involve in maintaining the integrity of the epithelial barrier by suppressing inflammation.Conclusion:The therapeutic effect of Aucklandia lappa Decne on Ulcerative colitis may be achieved by maintaining the integrity of the intestinal mucosal barrier and inhibiting inflammatory pathways...
