Cordyceps militaris is a kind of traditional mushroom that is medicinal and edible, itcontains many biological active substances such as cordycepin, cordycepins polysaccharideand cordycepic acid, and the cordycepin has many functions like anticancer, antibacterial,diminish inflammation and improve the immunity. The Cordyceps militaris culture mediumwere used as raw material, the extraction and separation of cordycepin and the process ofdecoloration of the extract liquid from Cordyceps militaris culture medium were studied inthis paper, with the purpose of make full use of the culture medium and lay the foundation ofdevelopment and utilization of cordycepin. The main research output was following:1. The influence of extraction temperature, extraction time, water material ratio and pHvalue to extraction yield were studied using quadratic general rotation unitized design method.The result showed that the influencing descending order of factors on the extraction yield waswater material ratio, extraction temperature, pH value and extraction time, by optimizing themodel, the optimum technological conditions were confirmed when extraction temperature,extraction time, water material ratio and pH value were63.3℃,4.8h,36.8and6.7.2. The resins suitable for separation of cordycepin were selected by static adsorption anddesorption experiments on the nine kinds of macroporous resins, and NKA-Ⅱresin wasfounded to be suitable for separation and purification of cordycepin, followed by LS-305resin.3. The NKA-Ⅱ macroporous resin was used to adorpt and desorpt cordycepin fromextract, and the key factors which influent the static adsorption and desorption effect werediscussed in this experiment. The result showed that in the process of static adsorption anddesorption on cordycepin, the optimum technological conditions by NKA-Ⅱ resin weredescribed as follows: Rotate speed, volume of sample, temperature, pH value and ethanolconcentration were155r/min,53mL,25℃,5.1and61%. The static adsorption anddesorption curves indicated that in the progress of adsorption, the appropriate adsorption timewas6hours, while during the progress of desorption, most of cordycepin was desorpted whenthe time reached180min. 4. In the process of dynamic adsorption and desorption on cordycepin, the optimumtechnological conditions by NKA-Ⅱr esin were described as follows: Loading flow rate was1.8BV/h in the progress of dynamic adsorption. Then use water to remove vestigialcomponent and impurities in the resin colume, its level is5folds the volume of the resin.During the progress of dynamic desorption, when the eluting rate pH value, ethanolconcentration, and volume of eluting regent were1.8BV/h,3.0,50%,5folds the volume ofthe resin, it has a central eluting peak as well as non-region streaking.5. The activated carbon was used to decoloring the extract from Cordyceps Militarismedium. In order to optium the technological conditions, the activated carbon dosage, timeand pH value were studied by using quadratic general rotation unitized design method on thebasis of single factor experiments.The decoloring effect was measured by was the colligationscore(T), the result showed that the influencing descending order of factors on the T wasactivated carbon dosage, pH value and time. By optimizing the model, the optimumtechnological conditions were confirmed when activated carbon dosage, time and pH valuewere1.0%,54min and3.5.
