Objective The aim of this study was to investigate the expression levels ofpigment epithelium-derived factor (PEDF) in different cell lines of nonsmall-cell lungcancer (NSCLC).Methods With immortalized lung epithelial cell line16HBE as control, forhuman lung adenocarcinoma cell line A549, squamous cell cancer cell line SK-MES-1and large cell carcinoma cell line NCI-H460, PEDF mRNA was assayed usingsemi-quantitative RT-PCR and PEDF protein in supernatant of24hours cell culture andwithin cell was detected by Enzyme-Linked Immunosorbent Assay (ELISA),respectively. Differences of PEDF levels between various groups were then analyzed.Results The expression levels of PEDF in the four cell lines(ie. A549,SK-MES-1, NCI-H460and16HBE) were increased by degrees in order: The relativeamounts of PEDF mRNA in A549, SK-MES-1, NCI-H460and16HBE were0.0226±0.0391,0.1001±0.09137,0.5489±0.1577and0.6856±0.0830, respectively;Accordingly, the PEDF protein levels of supernatant of24hours cell culture were(765.043±53.151) ng/L,(855.218±114.286) ng/L,(1240.465±36.5187) ng/L and(1290.359±19.981) ng/L, respectively; and the PEDF levels within cell protein were(640.344±95.370) ng/L,(719.520±15.517) ng/L,(997.071±105.512) ng/L and(1106.097±44.081) ng/L, respectively. Despite PEDF mRNA or PEDF protein level,excluding the difference between NCI-H460and16HBE as well as that between A549 and SK-MES-1, there was statistical difference between groups of A549and NCI-H460,A549and16HBE, SK-MES-1and NCI-H460as well as SK-MES-1and16HBE(P<0.05).Conclusions Regardless of the mRNA or protein level, PEDF may be lessexpressed in cell lines of lung adenocarcinoma and squamous cell lung cancercompared with large cell lung cancer or lung epithelial cell line; there seems nosubstantial difference between lung adenocarcinoma and squamous cell lung cancer aswell as large cell lung cancer and lung epithelial cell line. PEDF would thus hopefullybe of potentially diagnostic value in lung adenocarcinoma and squamous cell lungcancer, which deserves further clinical investigation.
