The Relationship Between Fecal Levels Of Calprotectin,tumor Dineric M2Type Pyruvate Kinase And Ulcerative Colitis | | Posted on:2014-02-20 | Degree:Master | Type:Thesis | | Country:China | Candidate:L Liu | Full Text:PDF | | GTID:2254330401470608 | Subject:Digestive science | | Abstract/Summary: | PDF Full Text Request | | ObjectiveIt was aimed to study the calprotectin and tumor M2type pyruvate kinase in fecal canbe used as active UC biomarkers and clinical value,through the detection of the fecal leveland expression in colon mucosa tissue of calprotectin and tumor M2type pyruvate kinasein patients with ulcerative colitis.MethodsAccording to the inclusion criteria,selected38controls and40patients with UC.According to the colitis activity index, the UC group was divided into2groups: activegroup28(cases) and inactive group12(cases).Enzyme linked immunosorbent assay wasused to determine calprotectin and tumor M2type pyruvate kinase in feces.Thestreptomyces avidin peroxidase of immunohistochemical technique was used to investigatethe expression of the calprotectin and tumor M2type pyruvate kinase in the colon mucosatissue.Results1.The fecal calprotectin concentration amid patients with active UC392.5(207~591.75μg/g) was significantly higher than that of inactive UC83.5(68.25~117.25μg/g)and controls23(19.5~27.75μg/g), and the level in active UC group was also higher thanthat control group P<0.001). The results also revealed a correlation between the fecalcalprotectin concentration and the endoscopic grades of UC (r=0.884,P<0.001).2.The fecal tumor tumor M2type pyruvate kinase concentration amid patients withactive UC40.5(9.13~81.25u/ml) was significantly higher than that of inactive UC7.9(3~ 9.68u/ml) and controls1.08(0.88~2.0u/ml), and the level in active UC group was alsohigher than that control group (P<0.001). The results also revealed a correlation betwixtthe fecal tumor M2type pyruvate kinase concentration and the endoscopic grades of UC(r=0.855,P<0.001).3.It was statistically significant with the comparison of Cal392.5(207~591.75μg/g)VS83.5(68.25~117.25μg/g), P<0.001and Tu M2-PK40.5(9.13~81.25u/ml) VS17(12~24.75u/ml), P<0.05in the active to the active group after treatment.4.There was a strong linear correlation between the levels of fecal Tu M2-PK with Calin UC (r=0.955P<0.01).Predetermined cut off level of53.0μg/g for fecal Cal and3.6u/mlfor Tu M2-PK test produced a sensitivity,specificity of80%and78%,86%and89%of thediagnosis UC.5.Different strength Cal and Tu M2-PK by immunohistochemical staining waspresented in the colon mucosa tissue of active UC,while only weak expression in inactiveUC and negative in controls.
