CASP3Gene Ploymorphism Associated With Kawasaki Disease And Coronary Artery Lesion

Objective:With the Kawasaki Disease (KD) group and health control group children’s genomic DNA in peripheral blood as the research object,and analyzing the base of caspase-3gene CASP3SNP rs113420705(formerly rs72689236),this study was conducted to discuss whether the CASP3SNP rs113420705is associated with Kawasaki disease or with Coronary artery lesions susceptibility in Chinese Han children,To provide reliable scientific basis for preventing the occurrence and early diagnosis of kawasaki disease and coronary artery leision.Methods:Choosing40patients with KD group and30healthy ethnically and gender-matched children with the control group in Tianjin children’s hospital from oct,2010to Apr,2012, and the genotype of the CASP3SNP rs113420705was detected by polymerase chain reaction (PCR) and the direct sequencing,analyzed the genotype of the rs113420705and investigated whether or not the the A allele frequency of the CASP3SNP rsl13420705is associated with the KD or KD with CAL susceptibility in Chinese Han children.Results:In this study, CASP3gene SNP rsl13420705site were detected in all subjects, there were three kinds of genotypes:AA、GA、GG.1. Between the Kawasaki disease group and health control group,the frequency distribution of the genotype (AA, GA, GG)(χ2=8.63, P<0.05) and the allele (χ2=9.03, P<0.05) showed significant statistically difference;And the A allele was risk factor of kawasaki disease in Chinese han children (OR=3.11).2. Between the group with Coronary artery lesions(CALs) and the group without Coronary artery lesions (NCALs), there was no significant statistically differences in the genotype frequency distribution (χ2=2.81, P>0.05) and the A allele frequency distribution (x2=2.21, P>0.05) of the CASP3gene rs113420705; but the A allele OR value was a risk factor of CAL in Chinese han kawasaki disease children (OR=1.97).Conclusions:1. The CASP3gene rs113420705SNPs gene polymorphism may be related to Chinese han children kawasaki disease.2. The A allele of CASP3gene rs113420705SNPs may be A risk factor for kawasaki disease in Chinese han children.3. The A alleles of CASP3gene rs113420705SNPs may be A risk factor for coronary artery leision in Chinese han children with kawasaki disease.
