Objective:The aim of this study is to identify the nuclear matrix proteins (NMPs) of BPH-1(benign prostatic hyperplasia cell), LNCaP (androgen dependent cell)and PC-3(Androgen independent cell) using two-dimensional electrophoresis coupled with mass spectrometry proteomics technology, which will pave the way for identification of PCa tissue samples.Methods:1, Three kinds of cells (BPH-1,LNCaP and PC-3) were cultured and the nuclear matrix proteins were extracted;2, NMPs were separated and identificated by two-dimensional electrophoresis and mass spectrometry;3, Western-blot were employed to explore the expression pattern of the identified proteins in prostate cancer cell lines.Results:1, Three kinds of cells were successfully cultured and extracted from nuclear matrix proteins in PCa cells. We find29differently expressed proteins and28of them were identified by mass spectrometry.2, three kinds of the proteins (hnRNPQ, Annexin A2and GRP78) were selected for further study with Western-blot. The results show that in extract of the cells, high levels of GRP78were found in BPH-1cell line. But the expression of GRP78in LNCaP and PC-3cell lines was very low or almost absent. Annexin A2was only expressed in PC-3cell and absent in BPH-1and LNCaP cell lines. No hnRNPQ expression was detected in cell lines.Conclusion:The expression of nuclear matrix proteins in three kinds of prostate cancer cell lines was significantly different. Studies have already confirmed that GRP78and Annexin A2are of great importance in cancer progress; they are likely to become candidate diagnostic biomarkers of prostate cancer; further more, GRP78and Annexin A2will play an important role on the mechanism of the development and progression of prostate cancer and provide a new target for treatment and diagnosis of cancer drugs, and have practical significance. Until now, little literature of hnRNPQ was publicly reported, so it may take time to give a full-fledged evaluation and prognosis. Further study is still needed in future.
