Objective:To establish the2-dimensional electrophoresis (2-DE) profiles of nuclear matrix proteins (NMPs) in human prostate cancer tissues and benign prostatic hyperplasia tissues; and to identificate the differentially expressed NMPs.Methods:1, Immunofluorescence technique was used to analysis the localization of proteins Annexin Ⅱ and GRP78in BPH-1、LNCaP and PC-3cell lines;2. Ammonium sulfate precipitation method was used to extract NMPs from tissues; The protein samples were separated by2-D DIGE;3.2-D electrophoresis maps was scanned by Typhoon9410multi-function imaging system;4. DeCyder5.0and MALDI-TOF was used to determine the differential expressions of proteins.Results:1. The expression of Annexin Ⅱ was very significantly higher in PC-3cells than in BPH-1cells and LNCaP cells lines. Annexin Ⅱ located in the cytoplasm and cell membrane of BPH-1, LNCaP and nucleus of PC-3;2. GRP78mainly located in the cytoplasm of BPH-1cells; and it was visible in nucleus and cytoplasm of LNCaP and PC-3cells;3. Obtained the map about the differential expressions of nuclear matrix proteins in human prostate cancer and human benign prostatic hyperplasia tissues;4.24differentially expressed proteins were successfully acquired including protease, enzymes, various cell proliferation factors etc.5. Eight NMPS were up-regulated and sixteen NMPs were down-regulated in prostate cancer tissues.Conclusion:1. The differential expressions of Annexin Ⅱ and GRP78maybe related with the development stages of prostate cancer cells;2. There were obvious differences of NMPs’expression profile between human prostate cancer and benign prostatic hyperplasia tissues.
