Objective Volatile anesthetics protect the myocardial from ischemia and reperfusion injury but the mechanisms for this protection are poorly understood. Caveolae, sarcolemmal invaginations, and caveolins, scaffolding proteins in caveolae, localize molecules involved in cardiac protection. To provide a theoretical basis for the specific mechanisms,we investigated the role of sevoflurane preconditioning on myocardial ischemia and reperfusion injury (I/R) and its effects on Caveolin-3.Methods The perfused models of Wistar rats’isolated heart were prepared by improved Langedorff perfusion device.Those models were randomly divided into three groups (n=10):normal control group (sham group), ischemia-reperfusion injury group (I/R group) and sevoflurane preconditioning+ischemia-reperfusion injury group (SP group),all of which had been perfused by K-H liquid for30minutes.The sham and I/R had went on been perfused for20minutes.While the SP had been perfused by the sevoflurane (2%) for15minutes first and the K-H for5minutes.Then I/R and SP had been made into global ischemia for40minutes and perfused for60minutes.The end of the balance perfusion,the moment of ischemic,the time of perfused at the30and60minutes.the heart rate (HR),left ventricular diastolic blood pressure (LVEDP),maximal rate of left ventricular pressure (+dp/dtmax) and maximal rate of decrease of left ventricular pressure (-dp/dtmax) were measured.At the60minutes of reperfusion.the myocardial tissue were taken from the apex and left ventricular to measure the Caveolin-3expression of myocardial cells.Results At the30and60minutes of reperfusion.the HR,LVHDP,+dp/dtmax and-dp/dtmax of I/R and SP had significantly reduced compared with the time the end of the balance perfusion and the moment of ischemic.While after perfused at the30and60minutes,the HR,LVEDP,+dp/dtmax and-dp/dtmax of the SP had significantly increased compared with the I/R.The expression levels of Caveolin-3protein did not change significantly,but the Caveolin-3distributed in the surface of the membrane of SP was significantly higher (P<0.01).Conclusion Sevoflurane preconditioning can effectively protect myocardial ischemia-reperfusion injury.And cardioprotective effects of sevoflurane pretreatment with caveolin-level film distribution increased.
