【Objective:】To Explore the methods for culture, identification of glioma stem cells and theeffect of the gamma knife treatment of glioma stem cells.【Method】The method, which is similar to the neural stem cells culture,is used to culturetumor stem cell and carried out the biochemical identification of self-renewal,proliferation anddifferentiation of cells. After Gamma knife treatment,cell proliferation and apoptosis changesare detected by cellular biochemistry, flow cytometry.【Results】Under the culture conditions of Serum-free medium, cancer stem cells are culturedderived from human glioma tissue. they have become sphericalsuspended growth and getthe proliferation and self-renewal capacity,expressing the CD133and nestin,containing theability to differentiate into astrocytes and neural elements. After gammaknife treatment,contrasting with the group of glioma cells,the number of glioma stem cells areable to survive for about more88/per field of view. The difference was statisticallysignificant (P <0.05). Before gamma knife treatment, about35%glioma stem cells combinedwith BrdU-positive cells, and it is about22%after the treatment, the difference was statisticallysignificant (P <0.05).The Glioma stem cells are apoptosis by even different dose of raysare afterthe treatment of gamma knife, but difference was statistically significant (P <0.05) between15Gy group and10Gy group.【Conclusion】under the conditions of serum-free medium, Human glioma tissue could culturethe cancer stem cells in. The treatment of gamma knife could inhibit proliferation of gliomastem cells, causing their apoptosis.Compared with glioma cells, glioma stem cells are notsensitive to gamma knife radiotherapy and Radioresistant.
