In order to reveal the change of the microspore during its development, I observed differentdevelopment periods of flower buds according to the optical microscope of the temperaturesensitive cytoplasmic male sterile line Y203. From May to October of2011and2012.Irecorded the change of the temperature (daily average temperature and the lowesttemperature and accumulated temperature) in order to set different temperatures in theincubators(24/18℃,31/27℃,Day/night),I just wanted to find the critical stage of fertilityconversion and the critical temperature.Changes in activity of POD and CAT and in content ofsoluble protein were investigated at different temperatures and different stages.At differentdevelopmental stages of temperature sensitive cytoplasmic male sterile line Y203, thesemi-quantitative determination had been done.The main results were obtained as follows:1.In order to find the critical temperature of temperature sensitive cytoplasmic malesterile line Y203,it is necessary to record the change of temperature in the field and to setdifferent temperatures of three incubators.2.Observing and comparing the different floral organs of temperature sensitivecytoplasmic male sterility pepper Y203at differernt temperature treatments,it shows the plantheight and stigma length of high-temperature treatment are higher than the low-temperaturetreatment plant.Observation of pollen germination rate at3,6,9h,21h,it was found inhigh-temperature materials for0, and low temperature materials with95%.3. There is no pollen in in the high-temperature treatment Y203,but the number of pollenin low-temperature treatmentY203is20457±1236.42,the pollen grain in it reach to91.31%±2.43%.4.Character analysis of different pepper materials and recording the plant height, stemdiameter, fruit weight, flesh thick, seed number, carpopodium length, fruit length, fruit settingrate, placenta length and width of temperature sensitive cytoplasmic male sterile line Y203、maintainer line HW203B、cytoplasmic male sterile line HW202A.5.Parafin section of the temperature sensitive cytoplasmic male sterile line Y203atdifferent temperature treatment shows high-temperature treatment was abortive, thedevelopment process of sporule at low temperature treatment is similar to Fertile plants.Themain reason of abortion is dued to early degradation of tapetal cells in high-temperaturetreatment Y203. 6.The contents of soluble protein in temperature sensitive cytoplasmic male sterile lineY203at different developmental stages and different treatments,the activity of POD and CATin different stages of temperature sensitive cytoplasmic male sterile line Y203has beenmensurated.7.Expression analysis of orf507in temperature sensitive cytoplasmic male sterile lineY203at different temperature treatments.
