With BUZHONG YIQI WAN residue as raw materials, this paper discussed the extraction process of insoluble dietary fiber by compound enzymolysis. The optimal conditions of dietary fiber which were extracted by compound enzymolysis methods was determined by single factor experiment, orthogonal design experiment, variance analysis, multiple comparisons, response surface analysis Method and the order of the influences of different factors was also researched. In addition, studies of dietary decolorization technology of dietary fiber and dietary fiber retention ability, oil-holding capacity, expansible capacity.(1) The technology of amylase enzymolysis:First, a single-factor test for concentration of amylase, pH, temperature, time, L/S and velocity of stirring was discussed, while the most influence factors of compound enzymolysis were screened by use of Plackett-Burman method. Followed the optimal condition of amylase enzymolysis:pH4.3,1.9%concentration of amylase,53.5℃,10/1L/S,200rpm of stirring,45min, optimized by response surface analysis Method. The removed efficiency of starch was82.1%in the technology.(2) The technology of neutral proteinase enzymolysis:With concentration of neutral proteinase, temperature, pH as the most influence factors, the optimal condition of neutral proteinase enzymolysis:3.5%concentration of neutral proteinase,55℃,45min, optimized by the method of quadratic regression orthogonal rotary. The hydrolysis of protein was above50%in the technology.(3) The technology of dietary fiber decolorization:With the whiteness as an evaluating indicator, the kinds of decolorant and conditon were studied by Ranking Method. The optimal condition of decolorization:H2O2as decolorant,6%concentration of H2O2, pH10、1h and20℃. The whiteness and taste of the dietary fiber were favorable, and the extraction rate of dietary fiber was61.2%.(4) The physical and chemical characteristics of dietary fiber extracted under optimal conditions were studied. Insoluble dietary fiber:the water holding capacity was 4.3g/g; the oil-holding capacity was3.2g/g; expansible capacity was5.3mL/mL; the cation exchanging capacity was0.76mmol/g; the adsorbing capacity of bile acids was0.045mg/g. The purity of product was79.65%.
